摘要
目的研究宫颈癌组织中MEG3及miR-9的表达情况,并探讨两者间的关联性。方法RNAhybrid软件预测MEG3与miR-9的结合位点。反转录实时荧光定量聚合酶联反应(RT-qPCR)检测宫颈癌组织中的MEG3及miR-9的表达,Kaplan-Meier生存分析评估预后。根据MEG3及miR-9表达量结果行Spearman相关分析;在宫颈癌细胞中过表达MEG3后检测miR-9的表达,研究MEG3对miR-9表达的影响。结果MEG3在宫颈癌组织中的表达较正常组织低(P<0.001);miR-9在宫颈癌组织中的表达较正常组织高(P<0.001)。MEG3低表达组、miR-9高表达组患者的中位生存时间及无瘤生存时间更短(P<0.05)。宫颈癌组织中的MEG3与miR-9的表达呈负相关(相关系数为-0.753,P<0.001);过表达MEG3后,HeLa、Siha细胞中miR-9表达均为下调(P<0.001)。结论MEG3与miR-9具有潜在的结合位点。宫颈癌组织中的MEG3呈低表达,miR-9呈高表达,MEG3低表达或miR-9高表达患者的预后较差。MEG3与miR-9的表达呈负相关性,在宫颈癌细胞株中MEG3过表达后能下调miR-9的表达,MEG3可能通过负向调节miR-9基因表达参与宫颈癌的发生发展。
Objective To investigate the expression of MEG3 and miR-9 in cervical cancer tissues and to explore the correlation.Methods The binding site of MEG3 and miR-9 was predicted by RNAhybrid software.The expression of MEG3 and miR-9 in cervical cancer tissues was detected by RT-qPCR,and the prognosis was evaluated by kaplan-meier survival analysis.Spearman correlation analysis was performed based on MEG3 and miR-9 expression results.After overexpression of MEG3 in cervical cancer cells,the expression of miR-9 was detected to study the influence of MEG3 on the expression of miR-9.Results The expression of MEG3 in cervical cancer tissues was lower than that in normal tissues(P<0.001).The expression of miR-9 in cervical cancer tissues was higher than normal tissues(P<0.001).The median survival time and tumor-free survival time of patients in the low-expression MEG3 group and the high-expression miR-9 group were shorter(P<0.05).MEG3 was negatively correlated with miR-9 expression in cervical cancer tissues(correlation coefficient was-0.753,P<0.001).After MEG3 overexpression,miR-9 expression in HeLa and Siha cells were down-regulated(P<0.001).Conclusions MEG3 has potential binding sites with miR-9.In cervical cancer tissues,MEG3 expression is low and miR-9 expression is high,and patients with low expression of MEG3 or high expression of miR-9 have poor prognosis.MEG3 is negatively correlated with the expression of miR-9.After overexpression of MEG3 in cervical cancer cell lines,the expression of miR-9 can be down-regulated,and MEG3 may be involved in the occurrence and development of cervical cancer by negatively regulating the expression of miR-9 gene.
