抗稻瘟病基因Pi9导入籼稻恢复系泉恢039的应用研究 被引量:1

摘要 【目的】对本单位育成的强优势恢复系泉恢039进行遗传改良,提高其稻瘟病抗性。【方法】用人工温汤去雄杂交法,以泉恢039为母本(受体亲本),与75-1-127(Pi9基因供体亲本)杂交,并用泉恢039作为轮回亲本,连续回交2次。回交后代先用系谱法结合叶片上的绒毛性状进行田间筛选,随后对入选株系在室内进行Pi9基因的分子检测和上杭县茶地镇田间自然诱发鉴定。入选株系同步用三系不育系191A和开源A进行恢复力检测。【结果】经田间筛选和室内分子标记检测,从36个具有绒毛性状的株系中,筛选出12个携带Pi9抗病等位基因,且株叶形态和恢复力与泉恢039基本一致的株系。经田间抗性鉴定,结果表明其中11个株系苗瘟抗性水平高于泉恢039,6个株系苗瘟抗性水平与对照75-1-127相当。【结论】利用Pi9基因和GL6绒毛基因同位于水稻第6染色体的遗传现象,可以通过以叶面绒毛等农艺性状进行田间筛选为主,简化室内Pi9基因检测频次和数量,从而选出携带Pi9抗病等位基因的株系(入选率为33.33%),为进一步筛选出符合育种目标的株系奠定基础。 【Objective】An Indica rice restorer line was genetically modified to improve the blast resistance.【Method】The Indica rice,Quanhui 039,formerly developed by the Institute of Agricultural Science was used as the female(recurrent)parent to cross with the Pi9-carrying 75-1-127 as the donor.The obtained F1 was backcrossed with Quanhui 039 twice,and the resulting progenies screened in the field by the pedigree method along with evidence of the leaf pubescence trait shown on the plants.The selected lines were confirmed by a positive detection of the molecular markers for Pi9 as well as the validation of blast resistance conducted at Chadi,Shanhang county.Furthermore,the lines were tested with CMS lines,191A and Kaiyan A,to verify their restore ability.【Result】In the 36 lines showing pubescence trait,12 carrying Pi9 as identified by field and marker-assisted selections had similar plant phenotype and restore ability as Quanhui 039.In the field,11 of the seedlings displayed greater blast resistance than Quanhui 039,and 6 equivalent to 75-1-127.【Conclusion】By using the presence of Pi9 in rice chromosome 6 and pubescence trait on the plant as the evidence,the current protocol considerably simplified the screening process of the genetically modified Indica rice lines with an accuracy rate as high as 33.33%.
