组胺在抽动障碍发病机制中的作用

Research progress on the role of histamine in the pathogenesis of tic disorder

摘要 组胺作为一种神经调节剂,与抽动障碍(TD)的关系正逐渐成为人们关注的焦点。自从在TD家系中发现组胺合成关键酶组氨酸脱羧酶(HDC)特定基因的突变,组胺参与TD发生的假说在人群及动物模型研究中得到了验证。组胺及其受体参与调节皮质-纹状体-丘脑-皮质(CSTC)环路、神经递质及小胶质细胞介导的神经炎症,影响TD的发生。组胺受体3(H3R)是TD很有前景的药物治疗靶点。本文综述了组胺及其受体在TD发病机制中的作用。 The relationship between histamine and tic disorder(TD)as a neuromodulator is gradually gaining attention.Specific gene mutations of histidine decarboxylase(HDC),a key enzyme in histamine synthesis,have been discovered in TD families,verifying the hypothesis that histamine is involved in TD generation in human and animal models.Histamine and its receptors play a role in regulating cortico-striato-thalamo-cortical(CSTC)circuits,neurotransmitters,and microglia-mediated neuroinflammation,which subsequently affect the development of TD.The H3receptor shows promise as a pharmacotherapeutic target for treating TD.This article reviews the role of histamine and its receptors in the pathogenesis of TD.
