摘要
应用超声波辅助水酶法提取菜籽油脂,应用响应面实验优化了超声参数,并在此基础上对比了不同酶作用下的提油率,同时研究不同酶对油脂脂肪酸分布的影响。选取了超声时间、超声温度、超声功率作为考察超声对细胞壁破坏效果的超声参数,并以提油率为考察指标,得到了最佳的超声参数:超声温度40℃、超声功率350 W、超声时间60 min,此时的提油率为67.55%。在此超声参数下,对比了Protex 6L、Protex 7L、Alcalase 2.4L、CellulaseViscozyme L五种酶对提油率的影响,其中Protex 6L、Protex 7L两种酶作用下提油率最高为72.87%、72.17%。经GC/MS分析油脂脂肪酸分布发现,不同酶作用下对油脂脂肪酸没有影响。
In the present study,the ultrasound-assisted enzymatic aqueous extraction of oil form rapeseeds was optimized using response surface methodology,and the effect of different enzymes on oil yield and fatty acids distribution was therefore investigated.Effects of sonication on breaking cell walls were evaluated based on analyzing ultrasonic time,ultrasonic temperature and ultrasonic power.Optimum ultrasonic conditions were ultrasonic temperature of 40℃,ultrasonic power of 350 W,and ultrasonic time of 60 min,and at this time the oil yield was 67.55%.Effects of five enzymes,Protex 6L,Protex 7L,Alcalase 2.4L,Cellulase and Viscozyme L on oil yield were evaluated using rapeseeds prepared under the optimum ultrasonic condition.Results showed that the highest oil yield of 72.87%and 72.17%was achieved using Protex 6L and Protex 7L.There was no difference between fatty acid distribution of oils extracted by using different enzymes.
