超声波辅助水酶法提取油茶籽蛋白中五种酶的比较及响应面优化的工艺的研究 被引量:1

摘要 以油茶籽为原料,采用水酶法结合超声波预处理技术提取油茶籽蛋白,在此基础上比较得出五种酶对油茶籽蛋白提取率的影响。在单因素基础上通过正交实验选出最佳的超声参数,然后采用最优超声参数,在料水比1∶6,酶解时间3.5 h,加酶量0.02%。在最适pH和温度下,油茶籽蛋白提取率由高到低依次为:Kemzyme>Viscozyme L>Protex 7L>Alcalase 2.4L>Protex 6L。在单因素实验基础上,采用响应面优化方法确定蛋白提取的最优条件,得到的最佳条件为:采用Kemzyme,料水比1∶5,酶解时间3.24 h,酶解温度68℃,pH 7.28,加酶量0.048%,响应面最优值为20.42%。 Considering camellia seed as a raw material,using aqueous enzymatic combination of ultrasonic pretreatment technology extracts camellia seed protein,the effect of five enzymes on the extraction rate of camellia seed protein's was studied.Selecting the optimal ultrasound parameters,based on the orthogonal experiment and using the optimal ultrasound parameters,at 1∶6 solids-to-liquid ratio,enzymatic hydrolysis time for 3.5 h,0.02%of the enzyme concentration,the optimum pH and temperature,camellia seed protein's extraction rate follows in descending order:Kemzyme>Viscozyme L>Protex 7L>Alcalase 2.4L>Protex 6L.Based on single factor experiment,response surface optimization method was used to determine the optimal condition for protein extraction.The result showed that Kemzyme at 1∶5 solids-to-liquid ratio,enzymatic hydrolysis time for 3.24 h,enzymatic hydrolysis temperature for 68℃,pH 7.28,amount of enzyme 0.048%,optimum value of response surface was 20.42%.
