一株产蛋白微藻Desmodesmus abundans ZM-4的单细胞分选、鉴定及对养猪废水降解特性研究

摘要 利用单细胞分选技术(PRECI SCS单细胞分选仪)在自然环境中分离、筛选出适合培养利用的高质量微藻ZM-4,并对其进行了鉴定、生长特性观察、理化性质测定和单因素实验分析,探究了其在不同养猪废水浓度、pH值、光照时间、光照强度、温度、碳氮比条件下对养猪废水降解能力的影响.结果表明:微藻ZM-4属于Desmodesmus abundans藻属,细胞内油脂产量、蛋白质产量、多糖产量分别为108.3,190.7,126.3 mg/g.根据单因素实验,微藻ZM-4对养猪废水降解能力的最优培养条件为:养猪废水COD质量浓度为800 mg/L,pH=7,光照时间为24h,光照强度为6000 lx,温度为30℃,碳氮比为8∶1. In this study,a high-quality microalgae ZM-4 was isolated by using single-cell sorting technology(PRECI SCS single-cell sorter)which was suitable for cultivation and utilization in the natural environment.The identification of species,growth characteristics,physical and chemical properties,and single-factor experimental research were carried out.To explore the effect of ZM-4 on the degradation capacity of piggery wastewater under different environmental factors,the single-factor experimental tests were carried out.The result showed that ZM-4 belonged to Desmodesmus abundans genus by 18 SrDNA sequencing.The results of physical and chemical properties showed that the intracellular oil production,protein yield and polysaccharide yield of ZM-4 were 108.3,190.7,126.3 mg/g,respectively.The obtained optimal cultural conditions were as follows:800 mg/L COD concentration,pH=7,light time of 24 h,light intensity of 6000 lx,temperature of 30℃,carbon-nitrogen ratio of 8∶1.The above research results can lay the foundation for the future application of microalgae to treat piggery wastewater and to produce biomass or energy.
