鄂产头顶一颗珠的HPLC指纹图谱及主成分分析 被引量:6

摘要 目的:建立土家族药物头顶一颗珠的HPLC指纹图谱检识方法,为头顶一颗珠的质量评价提供参考。方法:以Syncronis-C18柱(250mm×4.6mm,5μm)为色谱柱,采用乙腈-水为流动相进行梯度洗脱,测定湖北省不同产地头顶一颗珠的指纹图谱,筛选其共有模式,标定共有指纹峰,并对不同产地头顶一颗珠指纹图谱进行相似度比较和主成分分析。结果:鄂产头顶一颗珠指纹图谱含23个共有峰,经相似度评价,10批次的头顶一颗珠药材与共有模式比较相似度均达到0.90以上,整体相似度较好。经主成分分析,前5个主成分的累积方差贡献率为86.93%,10批样品的综合得分在0.175^-0.069之间,显示各批药材质量差异相对较小,其中,恩施市红土乡罗家乡产地药材综合得分最高,质量最好。结论:该研究建立的HPLC指纹图谱可作为头顶一颗珠药材质量鉴别依据,能有效评价鄂产头顶一颗珠药材的质量。 Objective:To establish a method for HPLC fingerprint identification and provide a reference for the identification of the authenticity of Trillium tschonoskii Maxim..Methods:Samples were separated by Syncronis-C18 column(250 mm×4.6 mm,5μm)with acetonitrile-water as gradient mobile phase.The fingerprints of Trillium tschonoskii Maxim.of different production areas in Hubei province were determined and the common patterns were screened.Chromatographic fingerprint similarity evaluation system and principal component analysis were used to analyze the data of Trillium tschonoskii Maxim.from different production areas.Results:A total of 23 common peaks was verified and the similarities of 10 batches of Trillium tschonoskii Maxim.from different producing areas were more than 0.90,and the overall similarity was good.Five principal components with the characteristic root cumulative contribution rate reaching 86.93%were screened out by PCA results.The comprehensive score of the 10 batches of samples was between 0.175 and-0.069,indicating that the difference in the quality of each batch of medicinal materials was relatively small,among which the Luojiaxiang,Hongtu Township of Enshi comprehensive score was the highest and the quality was the best.Conclusion:The established HPLC fingerprint can be used as the identification basis for Trillium tschonoskii Maxim.,which can effectively evaluate the quality difference of Trillium tschonoskii Maxim.native to different producing areas in Hubei.
