中药桃仁对糖尿病大血管纤维化大鼠TRIB3基因及Toll样受体信号通路的影响 被引量:10

Effects of peach kernel on TRIB3 gene and Toll like receptor signaling pathway in rats with diabetic macrovascular fibrosis

摘要 目的:观察糖尿病大血管纤维化大鼠模型中TRIB3 mRNA及Toll样受体信号通路表达及中药桃仁的影响。方法:雄性SD大鼠200只,随机选30只为空白组,余170只造糖尿病鼠模型,成功后随机选取40只为早期干预组,予桃仁颗粒剂水溶液10mL·kg-1·d-1灌胃,余进行糖尿病大血管纤维化造模,成模后分组:模型对照组、高剂量组、低剂量组;分别给予0.9%氯化钠溶液10mL·kg-1·d-1、桃仁颗粒剂水溶液20mL·kg-1·d-1和10mL·kg-1·d-1灌胃。干预第1、3、7周处死大鼠,检测TRIB3 mRNA及TLR4表达情况。结果:与模型对照组比较,第1、3、7周早期干预组TRIB3 mRNA及TLR4表达降低(P<0.05,P<0.01),第7周低剂量组、高剂量组TRIB3mRNA表达下降(P<0.05,P<0.01),第3、7周,低剂量组、高剂量组TLR4表达降低(P<0.01)。结论:TRIB3基因、TLR4和糖尿病大血管纤维化之间存在密切关系,桃仁可降低TRIB3 mRNA及Toll样受体信号通路表达。 Objective:To observe the expression of TRIB3 mRNA and Toll like receptor signal pathway in diabetic rats with macrovascular fibrosis and the effect of peach kernel.Methods:Two hundred male SD rats were randomly selected as the blank control group(30 rats)and 170 rats as the diabetic model(170 rats).Forty rats were randomly selected as the early intervention group(40 rats)after the success.The rats were given 10 mL·kg-1·d-1 of aqueous peach kernel granules to stomach,and the rest were given 10 mL·kg-1·d-1 of normal saline and 10 mL·kg-1·d-1 of peach kernel granules 20 mL·kg-1·d-1 and 10 mL·kg-1·d-1 of granule solution were gavage.Rats were killed at the 1 st,3 rd and 7 th week of intervention,and expression of TRIB3 mRNA and TLR4 were detected.Results:Compared with the model control group,the expression of TRIB3 mRNA decreased in the early intervention group(P<0.05,P<0.01)and in the low dose group and the high dose group(P<0.05,P<0.01)at the 7 th week.Compared with the model control group,TLR4 expression decreased in the early intervention group(P<0.05,P<0.01)and in the low dose group and high dose group(P<0.01)at the third and seventh weeks.Conclusion:There is a close relationship with diabetes major vascular fibrosis and TRIB3 mRNA and TLR4,peach kernel can reduce the expression of TRIB3 mRNA and Toll like receptor signal pathway.
