妊娠期高血压患者血清miR-218、miR-155和ET-1水平变化及其与病情严重程度之间的关系

Relationship of serum miR-218, miR-155 and ET-1 and the severity of the disease in patients with hypertensive disorder complicating pregnancy

目的:探索妊娠期高血压患者血清miR-218、miR-155和ET-1水平变化及其与病情严重程度之间的关系。方法:选择2018年6月-2020年6月本院收治的148例HDCP患者,根据病情严重程度分为单纯性妊娠期高血压(GHP,n=63)、轻度子痫前期(MPE,n=51)和重度子痫前期(SPE,n=34),另选择同期于本院体检的100例健康孕妇为对照组。检测各组收缩压(SBP)和舒张压(DBP),荧光定量PCR检测血清miR-218、miR-155水平,ELISA检测血清ET-1水平;Pearson相关性分析血清miR-218、miR-155和ET-1与HDCP患者疾病严重程度之间的相关性。结果:HDCP组患者的SBP、DBP、血清miR-155和ET-1水平高于对照组(P<0.05),血清miR-218水平低于对照组(P<0.05);GHP组患者血清miR-218水平高于MPE组和SPE组(P<0.05),血清miR-155和ET-1水平低于MPE组和SPE组(P<0.05);MPE组血清miR-218水平高于SPE组(P<0.05),血清miR-155和ET-1水平低于SPE组(P<0.05);血清miR-218与HDCP患者病情严重程度呈显著负相关(P<0.05),miR-155和ET-1与患者病情严重程度呈正相关(P<0.05);血清miR-218与miR-155、ET-1呈负相关关系(P<0.05),血清miR-155与ET-1呈正相关关系(P<0.05)。结论:血清miR-218、miR-155和ET-1水平水平变化与HDCP患者病情严重程度具有显著的相关性。

Objective:To explore the relationship between serum miR-218,miR-155 and endothelin-1(ET-1)levels and the severity of the disease in patients with hypertensive disorder complicating pregnancy(HDCP).Methods:A total of 148 HDCP patients who were admitted to the hospital from June 2018 to June 2020 were enrolled.According to different progression,they were divided into HDCP group(GHP,n=63),mild pre-eclampsia group(MPE,n=51)and severe pre-eclampsia group(SPE,n=34).Another 100 healthy pregnant women who underwent physical examination in the hospital during the same period were enrolled as control group.The systolic blood pressure(SBP)and diastolic blood pressure(DBP)in each group were detected.The levels of serum miR-218 and miR-155 were detected by fluorescence quantitative PCR,and serum ET-1 were detected by ELISA.The correlation between miR-218,miR-155,ET-1 and disease severity was analyzed by Pearson correlation analysis.Results:The levels of SBP,DBP,serum miR-155 and ET-1 in HDCP group were higher than those in control group(P<0.05),while levels of serum miR-218 were lower than those in control group(P<0.05).The level of serum miR-218 in GHP group was higher than that of MPE group and SPE group(P<0.05),and levels of miR-155 and ET-1 were lower than that of MPE group and SPE group(P<0.05).The level of serum miR-218 in the MPE group was higher than that in SPE group(P<0.05),while levels of miR-155 and ET-1 were lower than that in SPE group(P<0.05).Serum miR-218 was negatively correlated with the severity of HDCP(P<0.05),while miR-155 and ET-1 were positively correlated with the severity of HDCP(P<0.05).Serum miR-218 was negatively correlated with miR-218 and ET-1(P<0.05),serum miR-218 was positively correlated with ET-1(P<0.05).Conclusion:The changes of serum miR-218,miR-155 and ET-1 were significantly correlated with the severity of HDCP patients.
