调节性T细胞在子痫前期中的作用

摘要 目的:研究表明调节性T(Treg)细胞在母胎耐受和妊娠维持中具有重要作用。方法:收集2018年1月至2018年10月在广州医科大学附属广州市第一人民医院妇产科收治的PE患者(n=10)及正常孕妇(n=8)外周血、蜕膜和脐带血,并分离单个核细胞。流式细胞术检测外周血、蜕膜和脐带血中CD4^+CD25^+CD127^-的Treg细胞占CD4^+T细胞的比率,以及Treg细胞内TGF-β和IL-10的水平。结果:与健康孕妇相比,PE孕妇外周血及蜕膜的Treg细胞占CD4^+T细胞的比率显著降低(P<0.01,P<0.05),并且Treg细胞内TGF-β和IL-10的水平亦明显降低(P<0.05)。而PE孕妇的脐带血Treg比率及其细胞内TGF-β和IL-10的水平与健康孕妇无明显差异(P>0.05)。结论:系统和母胎界面Treg细胞比率和细胞内TGF-β和IL-10水平的下降可能在PE的发生中具有重要作用。 Objective:To determine the role of regulatory T(Treg)cells in the development of preeclampsia(PE).Methods:PE patients(n=10)and normal pregnant women(n=8)admitted to the Department of Obstetrics and Gynecology in Guangzhou First People’s Hospital affiliated to Guangzhou Medical University between January 2018 and October 2018 were enrolled.From these subjects,peripheral blood,decidua and umbilical cord blood were collected for separation of mononuclear cells.Flow cytometry was used to detect the ratio of CD4^+CD25^+CD127-Treg cells to CD4^+T cells in peripheral blood,decidua and cord blood,and the levels of TGF-βand IL-10 in Treg cells.Results:Compared with healthy pregnant women,pregnant women with PE showed significantly lower ratio of Treg cells to CD4^+T cells and lower levels of TGF-βand IL-10 in Treg cells in peripheral blood(P<0.01)and decidua(P<0.05).The PE patients and healthy pregnant women did not differ in the ratio of Treg cells to CD4^+T cells and the levels of TGF-βand IL-10 in Treg cells in umbilical cord blood(P>0.05).Conclusion:Reduction in ratio of Treg cells and levels of TGF-βand IL-10 in Treg cells in the circulation and maternal-fetal interface may play an important role in the occurrence of PE.
