‘西伯利亚’百合LiJAR1基因的克隆及表达分析 被引量:1

摘要 茉莉酸类物质(Jasmonates,JAs)是一类在植物体内广泛存在的激素,在植物次生代谢产物合成中起着重要的调控作用。花香作为植物重要的挥发性次生代谢产物,受到JAs信号的影响。为探究LiJAR1在百合单萜类挥发物合成中的作用,本研究以东方百合‘西伯利亚’(Lilium‘Siberia’)为试验材料,克隆LiJAR1基因,对其序列进行生物信息学分析,并通过荧光定量(RT-qPCR)技术进行时空表达分析。结果表明,LiJAR1基因全长1743 bp,编码580个氨基酸,具有典型的GH3特征结构域。LiJAR1相对表达量随着花朵的逐渐开放至盛开期而逐渐升高,转入衰败期开始降低。LiJAR1基因在内花被片中表达量最高,其次是叶和根,该基因在花柱、花丝、子房、花药中的相对表达量较低。本研究分析了LiJAR1基因的蛋白质结构功能,明确了该基因的表达模式,为后续研究其在百合单萜类挥发物释放中的信号作用奠定了基础。 Jasmonates(JAs)are a class of hormones widely found in plants,which play an important regulatory role in the biosynthesis of secondary metabolites.As the important plant secondary metabolites,floral volatiles are affected by JAs signal.In order to explore the role of LiJAR1 in the biosynthesis of monoterpenoid in lily,in this study,LiJAR1 gene in Lilium‘Siberia’was cloned,and bioinformatics analysis was carried out,and the temporal and spatial expression was also analyzed by RT-qPCR.The results showed that the amplified gene had a full length of 1743bp,and encoded 580 amino acids with a typical GH3 domain.The relative expression level of LiJAR1 increased gradually from flower opening to full bloom and began to decrease at the declining stage.The expression of LiJAR1 gene was the highest in inner tepals,followed by leaves and roots.The relative expression of LiJAR1 gene in style,filament,ovary and anther was low.In this study,the protein structure and function of LiJAR1 gene were analyzed,and the expression pattern of LiJAR1 gene was identified,which provided a foundation for the subsequent study on its signaling role in the biosynthesis of monoterpenoid volatiles in lily.
