双歧杆菌次级代谢产物生物合成基因簇挖掘与毒力因子分析

Gene Cluster Mining of Secondary Metabolite Biosynthesis and Virulence Factor Analysis in Bifidobacteria

摘要 为系统研究双歧杆菌(Bifidobacteria)次级代谢产物生物合成基因簇和毒力因子分布情况,本研究利用antiSMASH和PathoFact对1230株双歧杆菌全基因组序列进行了次级代谢产物生物合成基因簇和毒力因子预测分析。结果表明,25.77%的双歧杆菌基因组序列中含编码次级代谢产物生物合成基因簇,以编码合成羊毛硫肽化合物(lanthipeptide)、细菌素(bacteriocin)、杂胞型糖脂合酶样蛋白激酶(heterocyst glycolipid synthase-like protein kinase)和非核糖体多肽合成酶样片段(non-ribosomal peptide synthetase-like fragment)的基因簇为主。另外,在2株双歧杆菌基因组中预测发现了2个潜在的新次级代谢产物生物合成基因簇。毒力因子及毒素预测分析发现,绝大多数双歧杆菌基因组所编码的分泌型毒素种类较少,主要以假定的溶血素(hemolysin B)、核酸内切酶/核酸外切酶/磷酸酶家族(endonuclease/exonuclease/phosphatase family)以及肠毒素B(enterotoxin B)为主。生物合成基因簇的预测将为双歧杆菌次级代谢产物的制备和功能研究奠定基础,毒力因子分析将为双歧杆菌的生物安全性评价和有效开发等提供科学依据和参考。 To systematically study the distribution of secondary metabolite biosynthesis gene clusters and virulence factors in Bifidobacteria,the whole genome sequences of 1230 strains of Bifidobacterium were predicted and analyzed by using antiSMASH and PathoFact.The results showed that 25.77%of Bifidobacteria genome sequences contained gene clusters encoding secondary metabolite biosynthesis,mainly distributed in the types of lanthipeptide,bacteriocin,heterocyst glycolipid synthase-like protein kinase and non-ribosomal peptide synthetase-like fragment.In addition,two potential novel secondary metabolite biosynthesis gene clusters were found in the genomes of two Bifidobacterium strains.Virulence factor and toxin prediction analysis found that putative hemolysin,endonuclease/exonuclease/phosphatase family and enterotoxin B were the main types.The prediction of biosynthetic gene clusters will lay a foundation for the preparation and functional research of secondary metabolites of Bifidobacteria,and the virulence factor analysis will provide the scientific basis and reference for the biosafety evaluation and effective development of Bifidobacteria.
