摘要
本研究主要探讨Activin A对PCOS小鼠生殖功能的影响。将30只SPF级KM小鼠随机分为PCOS组,Activin A组和对照组,连续20 d皮下注射DHEA(6 mg/100 g/day)或油剂。第11 d和第16 d分别进行尾静脉注射Activin A(1 mg/kg)或生理盐水;第11~20 d每日对3组小鼠进行阴道涂片。实验结束后,取小鼠血清测定T、LH、E_(2)、P和FSH的水平;取卵巢进行病理组织学观察及LHR和FSHR mRNA的表达水平检测。结果表明,与PCOS组相比,Activin A组小鼠血清T、LH水平显著下降(P=0.013,P=0.038),E_(2)、FSH水平显著提高(P=0.049,P=0.036),P(P=0.069)水平差异不显著。LHRmRNA表达量显著下降(P=0.000),FSHRmRNA表达量显著提高(P=0.000);与对照组相比,PCOS组中小鼠血清T、P、LH水平显著升高(P=0.001,P=0.007,P=0.032),E_(2)、FSH显著下降(P=0.014,P=0.003);小鼠的发情周期和多囊性卵巢得到一定程度的改善。Activin A可改善小鼠生殖内分泌功能和动情周期,促进窦状卵泡的发育。
This study aims to explore the effect of Activin A on reproductive function of PCOS mice.In this research,thirty SPF KM mice were randomly divided into PCOS group,Activin A group and control group,and were subcutaneously injected with DHEA(6 mg/100 g/day)or oil solution for 20 consecutive days.Activin A(1 mg/kg)or normal saline was injected into the tail vein on day 11 and day 16,respectively.Vaginal smears were performed daily on day 11~20 in 3 groups of mice.At the end of the experiment,serum levels of T,LH,E_(2),P and FSH were measured.The ovaries were histopathologically observed and the expression levels of LHR and FSHR mRNA were detected.The results showed that compared with the PCOS group,testosterone and LH levels in the Activin A group were significantly decreased(P=0.013,P=0.038),E_(2)and FSH levels were significantly increased(P=0.049,P=0.036),and progesterone(P=0.069)levels were not significantly different.LHR mRNA expression significantly decreased(P=0.000),while FSHR mRNA expression significantly increased(P=0.000).Compared with the control group,testosterone,P and LH levels in the PCOS group were significantly increased(P=0.001,P=0.007,P=0.032),and E_(2),FSH were significantly decreased(P=0.014,P=0.003);the estrus cycle and polycystic ovary of mice were improved.Activin A can improve the reproductive endocrine function and estrus cycle of mice and promote the development of sinusoid follicles.
