急性髓系白血病患者LINC00324表达水平与染色体核型及预后的关系

Relationship between the Expression of LINC00324 and Karyotype and Prognosis in Patients with Acute Myeloid Leukemia

摘要 长链非编码RNA LINC00324在急性髓系白血病(acute myeloid leukemia,AML)发生发展中的作用尚不明确。为探讨AML患者LINC00324表达水平与染色体核型及预后的关系,首先采用实时荧光定量PCR法检测46例AML患者LINC00324的表达水平,并分析其与染色体核型及基因变化的关系;其次利用cBioPortal数据库分析173名AML患者LINC00324水平与各临床指标的关系。实验结果显示,AML患者LINC00324的表达水平明显降低(P<0.001),尤其在预后不良组显著下调(P<0.05);利用cBioPortal数据库分析也表明LINC00324水平在预后中等组和预后不良组中均显著下调(P<0.01),且预后不良组LINC00324水平更低(P<0.05)。在预后不良组中,单体核型患者LINC00324表达水平比非单体核型患者明显降低(P<0.05)。此外,LINC00324表达水平在FLT3-ITD阳性患者、M0、M1、M2型患者中明显下调(P<0.05);LINC00324水平与WT1表达量、C-KIT表达量和骨髓原始细胞数目均呈显著负相关(P<0.001)。LINC00324可能参与AML的发生和发展,有望作为AML患者预后评估的潜在指标。 The functional role of long-chain non-coding RNA LINC00324 in the genesis and development of acute myeloid leukemia(AML)remains unknown.To investigated the relationship between genome-wide expression level of LINC00324 and karyotype and prognosis in AML patients,firstly,real-time fluorescence quantitative PCR was carried out to detect the expression level of LINC00324 in 46 patients,and its relationship with chromosome karyotype and gene changes was analyzed.Secondly,cBioPortal database was utilized to analyze the relationship between LINC00324 expression level and clinical indicators in 173 AML patients.Experimental results exhibited that the expression level of LINC00324 was significantly reduced(P<0.001)in AML patients,particularly in the poor prognosis group(P<0.05);Data obtained from cBioPortal database also indicated that the level of LINC-00324 was significantly reduced in both the medium prognosis group and the poor prognosis group(P<0.01),and the poor prognosis group LINC00324 level was lower(P<0.05).Furthermore,in the group with poor prognosis,LINC00324 expression level in patients with monosomal karyotype was lower than that of patients with non-monosomal karyotype(P<0.05).Moreover,LINC00324 expression level was significantly down-regulated in FLT3-ITD positive patients,M0,M1,and M2 patients(P<0.05).Additionally,data also showed LINC00324 levels were negatively correlated with WT1 expression,C-KIT expression,and the number of bone marrow blasts(P<0.001).The results indicate that LINC00324 may be involved in the occurrence and development of AML,and is expected to be a potential indicator for the prognostic evaluation of AML patients.
