摘要
为了明确中国内蒙古呼和浩特市引起沙枣溃疡病的病原菌是否为葡萄蔓生小穴壳菌(Dothiorella viticola),本研究从典型的沙枣(Elaeagnus angustifolia L.)枝干发病组织中分离到了葡萄蔓生小穴壳菌,并应用科赫法则验证了该病原菌的致病性,同时结合形态特征和ITS(internal transcribed spacer)序列分析对病原菌进行鉴定。结果显示,沙枣溃疡病的病原菌形态特征与葡萄蔓生小穴壳菌相符,其ITS序列与已经公布的小穴壳菌的ITS相似度为100%,系统发育树分析结果与之一致。确认引起沙枣溃疡病的病原菌为葡萄蔓生小穴壳菌,该病害为沙枣上的国内新病害。
The aim of this study was to find out whether the main pathogen of Elaeagnus angustifolia canker in Hohhot city,Inner Mongolia,China is Dothiorella viticola.In this study,we isolated the D.viticola from the typical tissues of the branches of E.angustifolia.The pathogenicity of the pathogen was verified by Koch’s postulation,at the same time,the pathogen was identified by morphological characteristics and rDNA ITS sequence analysis.The results showed that the morphological characteristics of the pathogen of E.angustifolia canker were in accordance with those of D.viticola.The similarity between ITS sequence and ITS sequence of D.viticola that has been published is 100%,the result of phylogenetic tree analysis is consistent with that result.It is confirmed that the pathogen causing the stem canker of E.angustifolia is D.viticola,which is a new disease in China.
