摘要
miRNA作为进化保守的非编码RNA,对于细胞的发生、发展和增殖过程发挥重要的功能。miR-99b参与多种细胞的分化及增殖,但其在牛(Bas taurus)肌卫星细胞增殖分化中的表达以及可能靶向的基因尚不明确。本研究首先对牛基因组中miR-99b的基本信息进行详细介绍,采用茎环荧光定量PCR方法检测牛肌卫星细胞增殖及分化状态下miR-99b的表达。实验结果显示,miR-99b的表达在细胞处于增殖时显著高于其分化不同阶段。miRNA通过结合信使RNA转录本上的互补序列,转录完成后可以同时调节多个靶基因表达。为了深入研究miR-99b调节作用及机制,使用TargetScan软件预测了可能与miR-99b结合的靶基因,结果显示牛中大约有58个基因mRNA的3’UTR可能与miR-99b结合,并且这些靶基因大多数和细胞增殖、细胞分化及细胞凋亡相关。研究结果可为检测miR-99b在牛骨骼肌卫星细胞中的作用提供理论基础。
As an evolutionarily conserved non-coding RNA,miRNA plays an important role in the occurrence,development and proliferation of cells.miR-99 b is involved in the differentiation and proliferation of various cells,but its expression in the proliferation and differentiation of bovine muscle satellite cells and its possible target genes are unclear.This study first introduced the basic information of miR-99 b in the bovine genome,and stem-loop fluorescence quantitative PCR method was used to detect the expression of miR-99 b in the proliferation and differentiation state of bovine muscle satellite cells.The experimental results showed that the expression of miR-99 b during cell proliferation was significantly higher than its different stages of differentiation.The miRNA binds the complementary sequence on the messenger RNA transcript,which can simultaneously regulate the expression of multiple target genes after transcription.In order to further study the regulatory role and mechanism of miR-99 b,this study used TargetScan software to predict the target genes that may bind to miR-99 b.The results showed that the 3’UTR of about 58 gene mRNAs in cattle may bind to miR-99 b,and the majority of these target genes were relevant to cell proliferation,cell differentiation,and cell apoptosis.This study can provide theoretical basis for detecting the action of miR-99 b in bovine skeletal muscle satellite cells.
