砂藓(Racomitrium canescens)RcADH基因的克隆及生物信息学分析

摘要 本研究以砂藓(Racomitrium canescens)为材料,克隆得到砂藓乙醇脱氢酶基因RcADH。在拟南芥(Arabidopsis thaliana)中构建了RcADH过表达株系。通过对其生物信息学分析显示:RcADH的cDNA开放阅读框长1140 bp,编码含有379个氨基酸残基的多肽序列。RcADH蛋白理论等电点为6.79;脂肪指数为89.0,不稳定系数为24.35,属于不稳定的蛋白质。分析蛋白的跨膜结构域,结果显示RcADH基因编码的蛋白不含跨膜区。预测RcADH蛋白质中没有信号肽,属于非分泌型蛋白质。砂藓中RcADH蛋白与其它植物中的乙醇脱氢酶蛋白质序列具有较高的相似性,蛋白质亲缘关系系数为96.83%。本研究可为进一步探索RcADH基因功能提供基础。 In this study,Racomitrium canescens was used as experimental materia1,and an alcohol dehydrogenase gene RcADH was cloned from R.canescens by PCR.RcADH overexpression transgenic Arabidopsis thaliana was constructed.Bioinformatics analysis of RcADH showed that cDNA of RcADH was with the open reading frame of 1140 bp,a polypeptide sequence containing 379 amino acid residues was coded.For the RcADH protein,theoretical isoelectric point was 6.79;fat index was 89.0,instability index was 24.35,which was predicted that RcADH was an unstable protein.The transmembrane domain of the protein was analyzed,and the results showed that the protein encoded by RcADH gene had no transmembrane domain.It is predicted that there is no signal peptide in RcADH protein,which is a non-secreted protein.RcADH protein showed high similarity with alcohol dehydrogenase proteins in other plants,with the homologous index of 96.83%.The results in this study provides a basis for the further studies on the functions of RcADH.
