基于转录组测序的硒处理银杏萜类合成的关键基因表达分析 被引量:3

Transcriptome Sequencing Analysis of Gene Expression of Terpenoid Biosynthesis in Ginkgo biloba based on Selenium Treatment

摘要 本研究以银杏叶品种(Ginkgo biloba L.)家佛手室内盆栽为试材,对3年生银杏幼苗进行亚硒酸钠叶面喷洒处理,并分析其萜内酯相关基因的表达变化。挖掘与萜类化合物合成相关基因并对其差异表达基因进行筛选和分析,研究结果表明,亚硒酸钠处理叶片转录组测序共获得142834条Unigene,平均长度656 bp,基因差异表达(DEG)筛选得到5292个基因,并将其进行KOG功能分类。通过对萜类化合物合成相关KEGG途径进行分析,筛选出6条候选基因。采用实时荧光定量(qRT-PCR)对选取的候选差异基因进行验证,其中DXS、MECT、HMGR上调表达,与转录组测序结果一致,而DXR、MECPs、MVD在转录组测序中虽无明显差异,但q RT-PCR结果表明其在硒处理后的5周内就有明显差异变化。本实验为银杏提供了较为全面的基因信息和代谢途径数据,为硒处理条件下银杏分子调控机制的深入研究奠定基础。 Ginkgo biloba cultivar(Ginkgo biloba L.)was used as a test material in the study.The three-year-old ginkgo seedlings were sprayed with sodium selenite and the expression of azlactone and related genes were measured.The genes related to the synthesis of terpenoids were excavated and the differentially expressed genes were screened and analyzed.The results showed that 142834 Unigenes were obtained from the transcriptome sequencing of sodium selenite treatment,the average length was 656 bp,and the differential expression of genes(DEG)was screened.5292 genes were classified and KOG functionally classified.Six candidate genes were screened by analyzing the KEGG pathway involved in the synthesis of terpenoids.Real-time quantitative quantification(q RT-PCR)was used to verify the selected candidate differential genes.DXS,MECT and HMGR were up-regulated and consistent with the transcriptome sequencing results,while DXR,MECPs and MVD were not significantly different in transcriptome sequencing.However,qRT-PCR results showed significant differences in the 5 weeks after selenium treatment.This experiment provides a comprehensive information on genetic information and metabolic pathways for Ginkgo biloba,which lays a foundation for further research on the molecular regulation mechanism of Ginkgo biloba under selenium treatment conditions.
