摘要
为了揭示长期卵磷脂摄入搭配游泳能对高血压合并高脂大鼠的血压和脂代谢的影响。本研究将6周龄清洁级雄性SD大鼠建立高血压合并高血脂大鼠模型,每天按照2.0 g/kg的剂量对大鼠灌胃卵磷脂乳液(2 mL),共灌胃4周。大鼠每天进行30 min游泳运动,每周5次,共运动4周。给药和运动结束后,测量各组大鼠SBP和DBP以及血清TC、TG、LDL-c、HDL-c、MDA和SOD水平。苏木精-伊红(hematoxylin-eosin,HE)染色评价大鼠心肌组织病理变化。免疫组化检测大鼠心肌组织NF-κB、TGF-β1和TNF-α的阳性表达。Western blotting检测肝脏组织REBP-2、LDL-R、CYP7A1和HMGCR的表达。研究显示,卵磷脂联合游泳显著降低了高血压合并高血脂大鼠的SBP和DBP。卵磷脂联合游泳显著降低了大鼠的TC、TG和LDL-c水平并升高了HDL-c水平。卵磷脂联合游泳显著减轻了大鼠心肌组织病变。卵磷脂联合游泳显著降低了大鼠NF-κB、TGF-β1和TNF-α阳性染色评分。卵磷脂联合游泳显著升高了大鼠SOD水平并降低了MDA水平。卵磷脂联合游泳显著下调了大鼠肝脏组织中SREBP-2、LDL-R、CYP7A1和HMGCR的蛋白表达。本研究表明卵磷脂摄入搭配游泳具有较好的降血压和降血脂作用。卵磷脂和游泳的降血压和降血脂作用机制与改善体内氧化-抗氧化平衡、抑制炎症反应、调控SREBP-2信号通路有关。
In order to reveal the effects of long-term lecithin intake and swimming on blood pressure and lipid metabolism in hypertensive rats with high fat.In this study,a 6-week-old clean-grade male SD rat was established as a hypertensive and hyperlipidemic rat model.Rats were orally administered a lecithin emulsion(2 mL)at a dose of 2.0 g/kg for 4 weeks.The rats performed swimming for 30 min every day,5 times a week for 4 weeks.After administration and exercise,the levels of SBP and DBP and serum TC,TG,LDL-c,HDL-c,MDA and SOD in each group of rats were measured.Hematoxylin-eosin(HE)staining was used to evaluate the myocardial tissue pathological changes in rats.Immunohistochemistry was used to detect the positive expression of NF-κB,TGF-β1 and TNF-αin rat myocardial tissue.Western blotting was used to detect the expression of REBP-2,LDL-R,CYP7A1 and HMGCR in liver tissue.Studies showed that lecithin combined with swimming significantly reduced SBP and DBP in hypertensive rats with hyperlipidemia.Lecithin combined swimming significantly reduced TC,TG,and LDL-c levels and increased HDL-c levels in rats.Lecithin combined with swimming significantly reduced myocardial tissue lesions in rats.Lecithin combined with swimming significantly reduced the positive staining scores of NF-κB,TGF-β1 and TNF-αin rats.Lecithin combined swimming significantly increased SOD levels and decreased MDA levels in rats.Lecithin combined with swimming significantly down-regulated the protein expression of SREBP-2,LDL-R,CYP7A1 and HMGCR in rat liver tissue.This study showed that lecithin intake with swimming has a good effect on lowering blood pressure and blood lipids.The mechanism of lowering blood pressure and blood lipids of lecithin and swimming is related to improving the oxidation-antioxidation balance in the body,inhibiting the inflammatory response,and regulating the SREBP-2 signaling pathway.
