高浓度玛咖结合中强度跑步对2型糖尿病大鼠糖代谢和内分泌影响 被引量:2

Effects of High Concentration Maca Combined with Moderate Intensity Running on Glucose Metabolism and Endocrine in Type 2 Diabetic Rats

摘要 为了考察高浓度玛咖结合中强度跑步对2型糖尿病大鼠糖代谢和内分泌影响。本研究将60只SD大鼠随机分为对照组、模型组和干预组,每组20只。模型组和干预组大鼠通过注射60 mg/kg的STZ和高脂饲料喂养来建立2型糖尿病模型。干预组按照500 mg/kg·bw的剂量灌胃玛咖混悬液(2 m L)并进行中强度跑步运动进行干预,共干预4周。干预完成后分别检测各组大鼠血糖、肝糖原、肌糖原、血清INS、Lep、SOD、GSH-Px和MDA水平。苏木精和伊红(HE)染色检测大鼠胰腺组织形态学。免疫组化分析大鼠肾脏组织NF-κB、TGF-β1和TNF-α的阳性表达。Western blotting检测大鼠胰腺组织中GLUT-4和IRS-1的蛋白表达。研究显示,与模型组比较,干预组大鼠的FPG水平显著降低,而肝糖原和肌糖原含量显著升高。干预组的SOD和GSH-Px水平显著高于模型组,而MDA水平显著低于模型组。干预组的胰岛形状趋于正常,胰岛β细胞数量升高,形态基本正常。与模型组相比,干预组的NF-κB、TGF-β1和TNF-α染色评分均显著降低。干预组的INS和Lep水平显著低于模型组。干预组的GLUT-4和IRS-1的蛋白表达水平显著高于模型组。本研究表明,高浓度玛咖结合中强度跑步运动可有效降低2型糖尿病大鼠血糖水平并提高胰岛素敏感性。高浓度玛咖结合中强度跑步运动可能通过降低氧化应激损伤和炎症反应并激活GLUT-4-IRS-1信号来发挥治疗作用。 To investigate the effects of high concentration maca combined with moderate intensity running on glucose metabolism and endocrine in type 2 diabetic rats.Sixty SD rats were randomly divided into a control group,a model group and an intervention group,with 20 in each group.Model group and intervention group rats were injected with 60 mg/kg STZ and high-fat diet to establish type 2 diabetes model.The intervention group was administered with a maca suspension(2 mL)at a dose of 500 mg/kg·bw and intervened by medium-intensity running exercise for a total of 4 weeks.After the intervention,the blood glucose,liver glycogen,muscle glycogen,serum INS,Lep,SOD,GSH-Px and MDA levels of each group were measured.Hematoxylin and eosin(HE)staining were used to detect the morphology of rat pancreas.The expression of NF-κB,TGF-β1 and TNF-αin rat kidney was analyzed by immunohistochemistry.Western blotting was used to detect the expression of GLUT-4 and IRS-1 in rat pancreatic tissue.The study showed that compared with the model group,the FPG level of the rats in the intervention group was significantly reduced,while the levels of liver glycogen and muscle glycogen were significantly increased.The SOD and GSH-Px levels in the intervention group were significantly higher than those in the model group,while the MDA levels were significantly lower than in the model group.The shape of islets in the intervention group tended to be normal,the number of isletβcells increased,and the shape was basically normal.Compared with the model group,the NF-κB,TGF-β1 and TNF-αstaining scores in the intervention group were significantly reduced.The levels of INS and Lep in the intervention group were significantly lower than those in the model group.The expression levels of GLUT-4 and IRS-1 in the intervention group were significantly higher than those in the model group.This study shows that high-concentration maca combined with moderate-intensity running exercise can effectively reduce blood glucose levels and increase insulin sensitivity in type 2 diabetic rats.High concentration maca combined with moderate-intensity running exercise may play a therapeutic role by reducing oxidative stress injury and inflammatory response and activating the GLUT-4-IRS-1 signal.
