毛蕊花糖苷对力竭运动的骨骼肌损伤的保护作用 被引量:3

Protective Effect of Verbascoside on Skeletal Muscle Injury Induced by Exhaustive Exercise

摘要 毛蕊花糖苷是自然界双子叶植物中具有多种生理活性且普遍存在的重要物质之一,为了揭示毛蕊花糖苷对力竭运动所致骨骼肌损伤的保护作用及机制。本研究应用20 mg/kg·bw、50 mg/kg·bw和100 mg/kg·bw的剂量对SD大鼠灌胃毛蕊花糖苷溶液4周,然后进行力竭游泳实验。研究显示力竭运动组和毛蕊花糖苷组大鼠的血清葡萄糖(GLU)水平无显著差异,且均低于对照组。毛蕊花糖苷以剂量依赖方式降低了大鼠血清乳酸(Lac)和尿素氮(BUN)水平,而升高了游离脂肪酸(NEFA)水平。毛蕊花糖苷提高了大鼠肝糖原和肌糖原含量。毛蕊花糖苷升高了大鼠骨骼肌组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)水平并降低了丙二醛(MDA)水平。毛蕊花糖苷降低了大鼠血清肌酸激酶(CK)、天冬氨酸转氨酶(AST)和丙氨酸转氨酶(ALT)水平。苏木精和伊红染色显示,毛蕊花糖苷抑制了因力竭运动引起的骨骼肌纤维排列紊乱及炎性细胞浸润。由此表明毛蕊花糖苷一种具有较高开发价值的营养补充剂,可通过调节能量代谢方式、提高抗氧化能力、减少氧化应激损伤来减弱力竭游泳大鼠的骨骼肌损伤。 Verbascum glycosides are one of the important substances that have a variety of physiological activities and are ubiquitous in natural dicots.This study aims to reveal the protective effect and mechanism of verbascoside on skeletal muscle injury induced by exhaustive exercise.In this study,SD rats were given a solution of verbascoside glycosides for 4 weeks at doses of 20 mg/kg·bw,50 mg/kg·bw and 100 mg/kg·bw,followed by exhaustive swimming experiments.Studies showed that there was no significant difference in serum blood glucose(Glu)levels between the exhausted exercise group and the verbascoside group,and they were lower than the control group.Verbascoside reduced the levels of serum lactic acid(Lac)and urea nitrogen(BUN)in rats in a dosedependent manner,while increasing free fatty acids(NEFA)levels.Verbascoside increased rat liver glycogen and muscle glycogen content.Verbascoside increased the levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and catalase(CAT)in rat skeletal muscle tissue and decreased malondialdehyde(MDA)levels.Verbascoside reduced serum creatine kinase(CK),aspartate aminotransferase(AST)and alanine aminotransferase(ALT)levels in rats.Hematoxylin and eosin staining showed that verbascoside inhibited skeletal muscle fiber disorder and inflammatory cell infiltration caused by exhaustive exercise.It indicated that verbascoside is a nutritional supplement with high development value,which can attenuate skeletal muscle damage in exhausted swimming rats by regulating energy metabolism,increasing antioxidant capacity and reducing oxidative stress damage.
