运动疗法对肥胖大鼠的炎症反应和血脂代谢的影响

Effects of Exercise Therapy on Inflammatory Response and Lipid Metabolism in Obese Rats

摘要 为了揭示不同强度的离心运动对肥胖大鼠骨骼肌细胞凋亡和炎症的调节机制,本研究将80只雄性SD大鼠随机分为对照组、高脂饲料诱导肥胖组、低强度离心运动组(速度:15 m/min,时间:30 min)和高强度离心运动组(速度20 m/min,跑至力竭(>30 min)),每组20只,每周运动5次,共运动4周。采用苏木精-伊红(HE)染色检查大鼠骨骼肌形态;采用商用试剂盒检测大鼠骨骼肌氧化应激指标(SOD,CAT和MDA)水平;采用全自动生化分析仪检测TC和TG;采用ELISA试剂盒检测LDL和HDL;采用原位末端转移酶标记技术(TUNEL)检测骨骼肌细胞凋亡;使用Western blotting检测骨骼肌JNK、cleaved-caspase 3、Fas和Bcl-2的表达;使用ELISA试剂盒检测血清炎症因子(TNF-α,IL-1和IL-6)水平。研究显示,与肥胖组相比,低强度离心运动组的骨骼肌炎性细胞浸润显著减少,血清炎症因子(TNF-α,IL-1和IL-6)水平显著降低。与肥胖组相比,低强度离心运动组的SOD和CAT水平显著升高,而MDA水平显著降低;低强度离心运动组的骨骼肌细胞凋亡率显著降低;低强度离心运动组的JNK、cleaved-caspase 3和Fas的表达水平显著降低,而Bcl-2的表达水平显著升高。相反,高强度离心运动则会逆转上述指标的变化。离心运动组大鼠的TC、TG和LDL水平均显著低于肥胖组,而HDL水平显著高于肥胖组。本研究表明低强度离心运动可改善肥胖者体内的氧化-抗氧化平衡,抑制骨骼肌细胞凋亡和体内慢性炎症反应,并改善血脂代谢。 To reveal the mechanism of eccentric exercise with different intensity on skeletal muscle cell apoptosis and inflammation in obese rats.In this study,80 male SD rats were randomly divided into a control group,a high-fat diet-induced obesity group,a low-intensity eccentric exercise group(speed:15 m/min,time:30 min),and a high-intensity eccentric exercise group(speed 20 m/min,run to exhaustion(>30 min)),20 in each group,exercise5 times a week for a total of 4 weeks.Hematoxylin-eosin(HE)staining was used to examine the morphology of rat skeletal muscle.Commercial kits were used to detect oxidative stress indicators(SOD,CAT and MDA)in rat skeletal muscle.TC and TG were detected by automatic biochemical analyzer,and LDL and HDL were detected by ELISA kit.Skeletal muscle cell apoptosis was detected using in situ terminal transferase labeling technology(TUNEL).Western blot was used to detect the expression of JNK,cleaved-caspase 3,Fas and Bcl-2 in skeletal muscle.ELISA kit was used to detect serum inflammatory factors(TNF-α,IL-1 and IL-6).Studies showed that compared with the obese group,skeletal muscle inflammatory cell infiltration in the low-intensity eccentric exercise group was significantly reduced,and serum inflammatory factors(TNF-α,IL-1 and IL-6)levels were significantly reduced;the SOD and CAT levels in the low-intensity eccentric exercise group were significantly increased,while the MDA levels were significantly reduced;the skeletal muscle cell apoptosis rate in the low-intensity eccentric exercise group was significantly reduced;the expression levels of JNK,cleaved-caspase 3 and Fas in the low-intensity eccentric exercise group were significantly reduced,while the expression levels of Bcl-2 were significantly increased.On the contrary,high-intensity centrifugal motion reversed the changes of the above indicators.The levels of TC,TG and LDL in the centrifugal exercise group were significantly lower than those in the obesity group,while the levels of HDL were significantly higher than those in the obesity group.This study shows that low-intensity eccentric exercise can improve the oxidation-antioxidation balance in obese people,inhibit skeletal muscle cell apoptosis and chronic inflammatory response in the body,and improve blood lipid metabolism.
