基于Fe-MOFs的化学发光荧光双检测模式的免疫纸分析芯片的研究

摘要 采用Fe3+与2-氨基苯-1,4-二羧酸(NH2-BDC)室温下快速温和合成一种Fe-MOFs材料,此材料具有荧光及催化化学发光的性能,同时对于luminol-H2O2化学发光体系具有较强的催化能力。利用此MOFs材料表面的活性基团(-NH2),可将其作为信号探针标记于PSA的检测抗体上,采用荧光和化学发光双检测方式,在纸芯片上实现了血清中PSA的检测。其化学发光,荧光的线性范围分别为0.5~30和1.0~30 ng·mL-1,检出限分别为0.3和0.2 ng·mL-1。本研究工作利用MOFs材料建立了一种荧光、化学发光双检测模式的免疫纸分析芯片,丰富和拓展了MOFs材料在免疫纸芯片上的应用。 In this wok,a Fe-MOFs,consisting of Fe3+and 2-aminobenzene-1,4-dicarboxylic acid(NH2-BDC),was rapidly and mildly synthesized at room temperature.The Fe-MOFs showed a strong fluorescent signal and catalytic capacity for luminol-H2O2 chemiluminescence system.The active group—NH2 was beneficial for the connection of Fe-MOFs with the prostate specific antigen(PSA)detection antibody through the linker of glutaraldehyde.The Fe-MOFs was used as a signal antibody label for the detection of PSA on paper-based immune-devices to develop a paper-based chemiluminescence and fluorescence dual mode immunosensor for the detection of PSA.The PSA detection limits of chemiluminescence and fluorescence method were 0.3 and 0.2 ng·mL-1.The linear ranges were 0.5~30 and 1.0~30 ng·mL-1,respectively.In this work,a paper-based immune-device with fluorescence and chemiluminescence double detection mode was established by using MOFs materials.This strategy enriches and expands the development and application of MOFs materials in field of paper-based devices.
