摘要
目的探究延胡索提取物基于NOD样受体蛋白3(NALP3)炎性通路神经根型颈椎病大鼠的保护作用。方法用椎管插线法建立神经根型颈椎病模型,并随机分为模型组、阳性对照组、抑制剂组和低、中、高剂量实验组,每组16只。另取16只健康大鼠仅分离棘突两侧肌肉,剪断左侧C7椎板作为假手术组。假手术组和模型组均灌胃给予等体积0.9%NaCl;阳性对照组灌胃给予6.0g·kg^-1颈复康颗粒;抑制剂组给予尾静脉注射5μg·m L^-1Glybenclamide;低、中、高剂量实验组分别灌胃给予1.5,3.0和6.0 g·kg^-1延胡索提取物。6组大鼠每天给药1次,连续1周。用蛋白免疫印迹法检测NLRP3、白细胞介素-1β(IL-1β)、凋亡相关斑点样蛋白(ASC)和半胱氨酸蛋白酶1(Caspase1)蛋白的表达水平。结果低、中、高剂量实验组和假手术组、模型组、阳性对照组及抑制剂组的NLRP3蛋白表达水平分别为(0.72±0.14),(0.63±0.12),(0.42±0.16),(0.22±0.13),(1.09±0.16),(0.31±0.08)和(0.26±0.05),ASC蛋白表达水平分别为(0.71±0.09),(0.52±0.06),(0.38±0.07),(0.19±0.03),(0.87±0.08),(0.23±0.05)和(0.20±0.03),Caspase-l蛋白表达水平表达为(0.86±0.11),(0.73±0.14),(0.61±0.09),(0.17±0.03),(1.04±0.11),(0.36±0.07)和(0.23±0.04),IL-1β蛋白表达水平为(0.78±0.08),(0.54±0.07),(0.38±0.08),(0.16±0.04),(0.95±0.07),(0.26±0.03)和(0.21±0.04),模型组的上述指标与其余5组相比,差异均有统计学意义(均P<0.05)。结论延胡索提取物可通过抑制NALP3炎性通路,进而抑制神经炎性因子的表达,达到保护神经根型颈椎病大鼠神经根组织的作用。
Objective rydalis extract on the cervical spondylotic radiculopathy rats based on the inflammatory pathway of NOD-like receptor protein-3(NALP3).Methods lished by inserting spinal cord,and rats after modeling were randomly divided into model group,positive control group,inhibitor group and experimental-L,-M,-H groups with 16 cases per group.In addition,another 16 healthy rats were selected as sham operation group,for them only the muscles of both sides of spinous process were separated and the left C7 lamina was cut.The sham operation group and model group were given 0.9%NaCl by gavage.The positive control group was given 6.0 g·kg^-1 of Jingfukang granules by gavage.In the inhibitor group,5μg·m L^-1 of glybenclamide was injected into tail vein.1.5,3.0 and 6.0 g·kg^-1 Rhizoma Corydalis extract were given by gavage respectively in the experimental-L,-M,-H groups.Rats in the six groups were administrated once a day for one week.The expression levels of NLRP3,interleukin-1β(IL-1β),apoptosis related spot like protein(ASC)and Caspase-1 were detected by Western blot.Results positive control group and inhibitor group were(0.72±0.14),(0.63±0.12),(0.42±0.16),(0.22±0.13),(1.09±0.16),(0.31±0.08)and(0.26±0.05),the expression levels of ASC protein were(0.71±0.09),(0.52±0.06),(0.38±0.07),(0.19±0.03),(0.87±0.08),(0.23±0.05)and(0.20±0.03),the expression levels of Caspase-l were(0.86±0.11),(0.73±0.14),(0.61±0.09),(0.17±0.03),(1.04±0.11),(0.36±0.07)and(0.23±0.04),the expression levels of IL-1βprotein were(0.78±0.08),(0.54±0.07),(0.38±0.08),(0.16±0.04),(0.95±0.07),(0.26±0.03)and(0.21±0.04).Comparing with the other five groups,the above indexes in the model group were statistically significant(all P<0.05).Conclusion Corydalis extract can protect the nerve root tissue of rats with radiculopathy by inhibiting the inflammatory pathway of NALP3 and the expression of neuroinflammatory factors.
