枸杞子对非酒精性脂肪肝辅助性T细胞17和相关细胞因子的影响 被引量:12

Effects of Fructus Lycii on T helper cell 17 and related cytokines in non-alcoholic fatty liver disease

摘要 目的研究枸杞子对非酒精性脂肪肝(NAFLD)的辅助性T细胞17(Th17)及相关细胞因子的影响。方法按照随机数字表法将SD大鼠为4组:正常组、模型组、葛根组和枸杞子组,每组8只。用高脂饲料喂养8周制备NAFLD大鼠模型。枸杞子组和葛根组分别灌胃给予中药浓缩液0.5,1.5g·m L^-1,正常组和模型组给予0.9%Na Cl,每天1次,连续4周。检测各组大鼠总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、谷丙转氨酶(GPT)和谷草转氨酶(GOT)水平,以流式细胞仪检测外周血Th17、调节性T细胞(Treg)细胞含量,以实时定量聚合酶链反应法检测肝组织RORγt基因表达。结果模型组、枸杞子组和葛根组的TC水平分别为(7.87±0.31),(3.41±0.13),(3.23±0.14)mmol·L^-1;这3组的TG水平分别为(4.62±0.06),(1.40±0.24)和(1.49±0.33)mmol·L^-1;这3组的HDL-C水平分别为(0.92±0.12),(1.23±0.15)和(1.21±0.21)mmol·L^-1;这3组的LDL-C水平分别为(1.79±0.04),(0.48±0.12)和(0.51±0.22)mmol·L^-1;这3组的GPT水平为(102.4±5.1),(47.5±3.4)和(45.1±3.7)μ·L^-1;这3组的GOT水平为(175.8±3.3),(78.5±6.4)和(87.2±4.8)μ·L^-1;这3组的Treg/Th17为1.06±0.24,2.98±0.33和2.78±0.19;这3组的RORγt基因表达水平分别为2.16±0.14,1.45±0.32和1.56±0.12。上述指标:枸杞子组和葛根组与模型组相比,差异均有统计学意义(均P<0.05)。结论枸杞子能够通过抑制RORγt基因表达,在降低外周血Th17细胞含量的同时,升高Treg细胞含量,调整Th17/Treg平衡,进而抑制NAFLD发生发展。 Objective To study the effects of Fructus Lycii on T help cell 17(Th17)and related cytokines in nonalcoholic fatty liver(NAFLD).Methods The SD rats were randomly divided into 4 groups according to random number table:normal group,model group,puerarin group and Fructus Lycii group,with 8 rats in each group.Rats were fed with high-fat diet for 8 weeks for the preparation of NAFLD model.Rats in Fructus Lycii group and puerarin group were given intragastric administration of concentrated solution of traditional Chinese Medicine of the Fructus Lycii and puerarin 0.5,1.5 g·m L^-1,respectively;the model group and normal group was given normal saline,for 4 weeks.The levels of blood lipids including total cholesterol(TC),triglycerides(TG),low density lipoprotein-cholesterol(LDL-C),high density lipoprotein-cholesterol(HDL-C),glutamic pyruvic transaminase(GPT)and glutamic oxaloacetic transaminase(GOT)were measured.The contents of Th17 and regulatory T cell(Treg)in peripheral blood were detected by flow cytometry.The relative expression of RORγt gene in liver tissue was detected by real time quantitative polymerase chain reaction.Results The TC levels in model group,Fructus Lycii group and puerarin group were(7.87±0.31),(3.41±0.13),(3.23±0.14)mmol·L^-1,respectively;the TG in the three groups were(4.62±0.06),(1.40±0.24),(1.49±0.33)mmol·L^-1,respectively;the HDL-C in the three groups were(0.92±0.12),(1.23±0.15),(1.21±0.21)mmol·L^-1,respectively;the LDL-C in the three groups were(1.79±0.04),(0.48±0.12),(0.51±0.22)mmol·L^-1,respectively;the GPT in the three groups were(102.4±5.1),(47.5±3.4)and(45.1±3.7)μ·L^-1,respectively;the GOT in the three groups were(175.8±3.3),(78.5±6.4)and(87.2±4.8)μ·L^-1,respectively;the ratio of Treg/Th17 in the three groups were1.06±0.24,2.98±0.33,2.78±0.19,respectively;the relative expression of RORγt gene in the three groups were2.16±0.14,1.45±0.32,1.56±0.12,respectively.Comparing between Fructus Lycii group,puerarin group and model group,the differences of the factors were significant(all P<0.05).Conclusion Fructus Lycii can inhibit the expression of RORγt gene,decrease the content of Th17 cells in peripheral blood,increase the content of Treg cells,balance the ration of Th17/Treg,and inhibit the development of NAFLD.
