miR-212-3p靶向长链非编码RNA SCAMP1在胃癌中的差异表达

Different expressions of miR-212-3p associated targeting LncRNA SCAMP1 in Gastric cancer

目的:探究miR-212-3p靶向LncRNA SCAMP1在胃癌中表达水平。方法:通过生物信息学分析,预测miR-212-3p靶向的LncRNA;运用COX风险模型,分析miR-212-3p相关的靶向LncRNA SCAMP1对胃癌患者生存时间的影响;采用实时定量PCR,检测人胃癌组织及远癌组织中miR-212-3p和LncRNA的表达水平。结果:LncRNA预测结果显示miR-212-3p靶向SCAMP1等21个LncRNA;COX风险模型显示miR-212-3p在胃癌中无统计学差异(P>0.05),SCAMP1在胃癌中有统计学差异(P<0.05),其表达升高不利于患者预后;实时定量PCR结果显示,miR-212-3p在远癌组织表达量高于胃癌组织(P<0.05);SCAMP1在远癌组织表达量低于胃癌组织(P<0.05)。结论:miR-212-3p相关的靶向LncRNA SCAMP1高表达不利于胃癌患者的预后。

Objective:To investigate the expression of miR-212-3 p associated targeting LncRNA SCAMP1 in Gastric cancer.Methods:miR-212-3 p associated targeting LncRNA was predicted by starBase 2.0.COX Risk model was used to analyze the effects of miR-212-3 p associated targeting SCAMP1 on the survival time of gastric cancer patients.And real-time quantitative PCR was used to detect the expression levels of miR-212-3 p and LncRNA in human gastric cancer tissues and distant cancer tissues.Result:Prediction results of LncRNA showed that there were 21 LncRNAs including SCAMP1 which were targeted by miR-212-3 p.The result of COX risk model showed that there were no significant differences of the expressions of miR-212-3 p in gastric cancer and distant cancer tissue(P>0.05),while the expressions of SCAMP1 were significantly different in gastric cancer and distant cancer tissue(P<0.05).For the increased expression of miR-212-3 p was not good for the prognosis of gastric cancer patients.Real-time PCR analysis verified that miR-212-3 p was more highly expressed in distant cancer tissue than in gastric carcinoma(P<0.05),while SCAMP1 was more lowly expressed distant cancer tissue than in gastric carcinoma(P<0.05).Conclusion:The high expression of miR-212-3 p associated targeting LncRNA SCAMP1 may be unfavorable for the prognosis of gastric tumor patients.
