转谷氨酰胺酶催化增强明胶基粘合剂 被引量:2

Transglutaminase Enhanced Gelatin-based Adhesives

摘要 明胶基粘合剂因其良好的生物相容性被广泛的应用于组织工程领域,但其粘合能力仍存在较大提升空间。在本研究中,通过葡聚糖醛与明胶间的席夫碱反应形成一层交联网络,同时加入转谷氨酰胺酶,使其与明胶利用酶促交联反应形成另一层交联网络,通过双交联网络的形成提高了明胶基粘合剂性能。通过小瓶倒置、溶胀、压缩和搭接剪切等测试凝胶性能及粘合剂性能。结果表明,改性后的明胶基粘合剂具有适宜注射使用的凝胶时间,较低的溶胀比,压缩模量最高可达33.6kPa,是未改性明胶的4倍,粘合强度最高可达25.9kPa,是未改性明胶的12倍。明胶基粘合剂的性能得到了提升。 Because of their good biocompatibility,gelatin-based adhesives are widely used in the field of tissue engineering,but their adhesive ability still needs to be improved.In this paper,the oxidized dextran and gelatin formed one cross-linking network through Schiff base reaction,while enzymatic cross-linking reaction between transglutaminase and gelatin formed another cross-linking network.The formation of the double cross-linking network can enhance the bulk strength of gelatin adhesive.The properties of the adhesive were characterized by vial inversion,swelling,compression,lap shear,etc.The modified gelatin-based adhesive’s gelation time was suitable for injection,and the obtained gel showed a lower swelling ratio.The adhesive’s compression modulus was up to 33.6 kPa,which is 4 times that of unmodified gelatin.And the bonding strength of adhesive was up to 25.9 kPa,it is 12 times that of unmodified gelatin.The performance of gelatin-based adhesives has been improved.
