WHSC1L1表达与三阴性乳腺癌患者临床病理特征的关系及其潜在机制 被引量:2

The relationship between WHSC1L1 expression and clinicopathological features of triple-negative breast cancer patients and its underlying mechanism

摘要 目的探讨Wolf-Hirschhorn综合征候选基因1样1(WHSC1L1)三阴性乳腺癌(TNBC)中的表达及其潜在机制。方法收集我院收治的77例TNBC患者的癌组织和癌旁组织,免疫组织化学法、qRT-PCR法检测组织中WHSC1L1的表达。分析WHSC1L1的表达和患者临床病理资料的关系。细胞实验中,干预TNBC细胞中WHSC1L1的表达,MTT检测各组细胞增殖活力,Transwell实验检测各组细胞侵袭。检测TNBC细胞中ERK通路相关因子的表达,验证WHSC1L1通过ERK通路对TNBC进行调控。结果免疫组化检测结果显示,相对于癌旁组织,癌组织中WHSC1L1的阳性表达增强。此外,q RT-PCR检测结果也显示癌组织中WHSC1L1表达增强(t=12.91,P<0.001)。WHSC1L1的高表达与TNBC患者的肿瘤大小(P=0.026)和组织学分级(P=0.033)有关。与si-NC组比较,敲减WHSC1L1能够抑制TNBC细胞的增殖活力和侵袭。过表达WHSC1L1能够激活ERK信号通路。WHSC1L1对TNBC增殖和侵袭的影响可能是通过调控ERK信号通路实现的。结论WHSC1L1在TNBC中高表达,敲减WHSC1L1能够抑制TNBC细胞增殖和侵袭,该作用可能是通过调控ERK通路实现的。 Objective To investigate the expression and potential mechanism of histone methyltransferase WHSC1L1 in triple negative breast cancer(TNBC).Methods The cancer tissues and adjacent tissues of 77 TNBC patients admitted to our hospital were collected,and the expression of WHSC1L1 in the tissues was detected by immunohistochemistry and qRT-PCR.The relationship between the expression of WHSC1L1 and the clinicopathological data of patients was analyzed.In cell experiments,the expression of WHSC1L1 in TNBC cells was interfered,MTT was used to detect cell proliferation activity in each group,and Transwell assay was used to detect cell invasion in each group.The expression of ERK pathway-related factors in TNBC cells was detected to verify that WHSC1L1 regulates TNBC through the ERK signaling pathway.Results Immunohistochemical detection showed that the positive expression of WHSC1L1 in cancer tissues was enhanced compared with that in adjacent tissues.In addition,qRT-PCR detection results also showed that the expression of WHSC1L1was enhanced in cancer tissues(t=12.91,P<0.001).High expression of WHSC1L1 was associated with tumor size(P=0.026)and histological grade(P=0.033)in TNBC patients.Compared with si-NC group,knockdown of WHSC1L1 could inhibit the proliferation and invasion of TNBC cells.Overexpression of WHSC1L1 could activate the ERK signaling pathway.The effects of WHSC1L1 on TNBC proliferation and invasion may be achieved by regulating the ERK signaling pathway.Conclusion WHSC1L1 is highly expressed in TNBC.Knockdown of WHSC1L1 can inhibit proliferation and invasion of TNBC cells,which may be achieved by regulating the ERK pathway.
