痰瘀互结型冠心病大鼠模型中大鼠血清中hs-CRP、TNF-α与血脂水平相关性分析 被引量:2

摘要 目的建立痰瘀互结证型冠心病大鼠模型,分析模型大鼠血清超敏C-反应蛋白(hs-CRP)、肿瘤坏死因子-α(TNF-α)和血脂的相关性探讨血脂水平对模型大鼠冠心病发生发展的影响。方法通过对实验大鼠进行高脂饲料喂养和冠状动脉左前降支结扎建立痰瘀互结证型冠心病大鼠模型,喂养56 d后称量大鼠体质量,采用苏木精-伊红(HE)染色对大鼠心脏进行观察。检验大鼠腹主动脉血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)和血清hs-CRP、TNF-α的水平。对大鼠hs-CRP和TNF-α与血脂水平进行相关性分析。结果痰瘀互结证型冠心病大鼠的冠状动脉血管管腔明显狭窄,可见脂质堆积且出现粥样硬化,同时可见散在的心肌梗死灶。随着高脂饲料喂养时间的增加大鼠体质量、hs-CRP以及TNF-α水平呈现显著增加的趋势。各组大鼠血清TG水平差异无统计学意义(P>0.05),随着大鼠采用高脂饲料时间的增加TC和LDL的水平呈现显著增加的趋势HDL的水平呈现显著降低的趋势。未采用高脂饲料喂养的大鼠的血清hs-CRP、TNF-α和TC、TG、LDL、HDL的相关性均未出现统计学意义(P>0.05);高脂饲料喂养的大鼠的血清hs-CRP和TC、LDL均呈现显著正相关(P<0.05),与HDL均呈现显著负相关(P<0.05)。高脂饲料喂养的大鼠的血清TNF-α和TC、TG和LDL均呈现显著正相关(P<0.05),与HDL均呈现显著负相关(P<0.05)。结论痰瘀互结证型冠心病大鼠模型中,大鼠的血脂水平能够影响大鼠冠心病发展。 Objective To establish a rat model of Tan Yu Hu Jie type coronary heart disease(CHD),analysis the correlation between serum high-sensitivity C-reactive protein(hs-CRP),Tumor necrosis factor(TNF-α)and blood lipids in model rats and explore the effect of blood lipid level on the occurrence and development of CHD in model rats.Methods Establishment of a rat model of Tan Yu Hu Jie type CHD by high-fat diet feeding and left anterior descending coronary artery ligation,weighted rat after 56 days of feeding,observed the rat heart by HE staining,test the levels of serum total cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL),high density lipoprotein(HDL)and hs-CRP,TNF-αin rat abdominal aorta.Analysis the correlation between hs-CRP,TNF-αand blood lipid levels in model rats.Results The Tan Yu Hu Jie type CHD rat’s coronary lumen was significantly narrowed,and lipid accumulation and atherosclerosis were observed,and scattered myocardial infarction was observed.The body weight,hs-CRP and TNF-αlevels of rats increased significantly with the increase of feeding time of high-fat diet.There was no difference in serum TG levels between the groups(P>0.05),as the increase of the time of high-fat diet in rats,the level of TC and LDL showed a increase trend and the level of HDL showed a decrease trend.Serum TC,TG,LDL,HDL and hs-CRP,TNF-αshown no statistically correlation in rats without fed high-fat diet(P<0.05).There was a significant positive correlation between serum hs-CRP and TC,LDL in rats fed with high-fat diet(P<0.05),and a significant negative correlation with HDL(P<0.05).There was a significant positive correlation between serum TNF-αand TC,TG and LDL in rats fed with high-fat diet(P<0.05),and a significant negative correlation with HDL(P<0.05).Conclusion In the rat model of Tan Yu Hu Jie type CHD,the blood lipid levels can affect the development of coronary heart disease in rats.
