脑卒中后抑郁大鼠海马区BDNF表达与神经行为的相关性 被引量:5

摘要 目的探讨脑卒中后抑郁大鼠海马区脑源性神经营养因子(BDNF)表达与神经行为的相关性。方法60只大鼠随机均分为模型组与对照组,模型组建立了脑卒中后抑郁模型,对照组以假手术代替。记录大鼠的造模情况与神经行为状况,检测海马区BDNF表达水平并进行相关性分析。结果对照组在实验过程中无死亡情况,模型组建模成功25例,成功率为83.3%;模型组大鼠造模后7 d、28 d的神经功能缺损评分都高于对照组,对比差异均有统计学意义(P<0.05);模型组大鼠造模后7 d、28 d的体质量都显著低于对照组(P<0.05),模型组大鼠造模后7 d、28 d的血清白介素(IL)-6与C-反应蛋白(CRP)值都显著高于对照组(P<0.05),模型组大鼠造模后78 d的海马区BDNF相对表达水平显著低于对照组(P<0.05);模型组大鼠海马区BDNF相对表达水平与神经功能缺损评分存在负相关性(r=-0.562,P=0.000)。结论脑卒中后抑郁大鼠海马区BDNF表达水平呈现下降的趋势,也伴随有炎症因子的大量释放,与大鼠的神经行为有显著相关性。 Objective To explore the correlation between brain-derived neurotrophic factor(BDNF)expression and neurobehavior in hippocampus of post-stroke depression rats.Methods Sixty cases of rats were randomly divided into two groups-model group and control group.Post-stroke depression model were established in the model group,and no model were established in the control group.The modeling and neurobehavioral status of rats were recorded,and the expression level of BDNF in hippocampus were detected and were given correlation analysis.Results There were no deaths in the control group during the experiment.The model group were successfully modeled in 25 cases with the success rates of 83.3%.The neurological deficit scores of the model group at 7 and 28 days after the model were higher than those of the control group(P<0.05);The body weight of the rats in the model group were significantly lower than that of the control group at 7 days and 28 days(P<0.05),and the serum interleukin(IL)-6 and C-reactive protein(CRP)values of the rats in the model group were were significantly higher than those in the control group(P<0.05),and the relative expression levels of BDNF in the hippocampus of the model group at 78 days after modeling were significantly lower than those in the control group(P<0.05);There were negative correlation between the relative expression level of BDNF in hippocampus and neurological deficit score in the rats of the model group(r=-0.562,P=0.000).Conclusion The expression level of BDNF in hippocampus of depressive rats after stroke shows a downward trend,which is accompanied by a large amount of inflammatory factors released,which is significantly related to the neurobehavior of rats.
