仙灵骨葆胶囊对骨质疏松症大鼠BMP-2、BMP-4、TGF-β1和PKC蛋白表达影响 被引量:7

摘要 目的探讨仙灵骨葆胶囊对骨质疏松症大鼠骨形成蛋白-2(BMP-2)、骨形成蛋白-4(BMP-4)、转化生长因子β1(TGF-β1)和蛋白激酶C(PKC)蛋白表达影响。方法40只SD大鼠随机分为假手术组、模型组、对照组和实验组,每组大鼠10只。假手术组给予等剂量生理盐水灌胃;模型组:给予等剂量生理盐水灌胃;对照组:给予盖天力牡蛎钙0.3 g/kg灌胃;实验组:给予仙灵骨葆胶囊0.4 g/kg灌胃。各组大鼠灌胃均为每日1次,连续灌胃8周。采用双能X线骨密度仪检测股骨和腰椎骨密度;采用ELISA法检测血清Ca和P含量;采用免疫组化法检测骨组织BMP-2、BMP-4、TGF-β1和PKC蛋白表达。结果除假手术组,其余各组大鼠均造模成功。模型组、对照组、实验组股骨和腰椎骨密度低于假手术组(P<0.05);对照组、实验组股骨和腰椎骨密度高于模型组(P<0.05);实验组股骨和腰椎骨密度高于对照组(P<0.05)。模型组、对照组、实验组血清Ca和P含量低于假手术组(P<0.05);对照组、实验组血清Ca和P含量高于模型组(P<0.05);实验组血清Ca和P含量高于对照组(P<0.05)。模型组、对照组、实验组BMP-2、BMP-4和PKC蛋白表达高于假手术组,而TGF-β1蛋白表达低于假手术组(P<0.05);对照组、实验组BMP-2、BMP-4和PKC蛋白表达低于模型组,而TGF-β1蛋白表达高于假手术组(P<0.05);实验组BMP-2、BMP-4和PKC蛋白表达低于对照组,而TGF-β1蛋白表达高于对照组(P<0.05)。结论仙灵骨葆胶囊可增加骨质疏松症大鼠骨密度,认为可能与下调BMP-2、BMP-4和PKC蛋白表达及上调TGF-β1蛋白表达相关。 Objective To investigate the effects of Xianling gubao capsule on the expression of bone morphogenetic protein-2(BMP-2),bone morphogenetic protein-4(BMP-4),transforming growth factor-β1(TGF-β1)and protein kinase C(PKC)in osteoporotic rats.Methods The 40 SD rats were randomly divided into sham operation group,model group,control group and experimental group,10 rats in each group.Sham operation group was given equal dosage of normal saline;model group was given equal dosage of normal saline;control group was given 0.3 g/kg of calcium of Gaitianli oyster;experimental group was given 0.4 g/kg of Xianlinggubao capsule.Rats in each group were intragastrically administered once a day for 8 weeks.Bone mineral density of femur and lumbar spine was measured by dual-energy X-ray absorptiometry,serum Ca and P were measured by ELISA,BMP-2,BMP-4,TGF-β1 and PKC were detected by immunohistochemistry.Results Except for the sham operation group,the other groups were successfully established.Bone mineral density of femur and lumbar spine in model group,control group and experimental group was lower than that in sham operation group(P<0.05);bone mineral density of femur and lumbar spine in control group and experimental group was higher than that in model group(P<0.05);bone mineral density of femur and lumbar spine in experimental group was higher than that in control group(P<0.05).The levels of serum Ca and P in model group,control group and experimental group were lower than those in sham operation group(P<0.05);the levels of serum Ca and P in control group and experimental group were higher than those in model group(P<0.05);and the levels of serum Ca and P in experimental group were higher than those in control group(P<0.05).The expression of BMP-2,BMP-4 and PKC in model group,control group and experimental group was higher than that in sham operation group,while the expression of TGF-β1 was lower than that in sham operation group(P<0.05);the expression of BMP-2,BMP-4 and PKC in control group and experimental group was lower than that in model group,while the expression of TGF-β1 was higher than that in sham operation group(P<0.05);the expression of BMP-2,BMP-4 and PKC in experimental group was lower than that in control group,while the expression of TGF-β1 was higher the control group(P<0.05).Conclusion Xianling gubao capsule can increase bone mineral density in osteoporotic rats,which may be related to down-regulation of BMP-2,BMP-4 and PKC protein expression and up-regulation of TGF-β1 protein expression.
