青藤碱体内外微透析回收率的研究 被引量:1

摘要 目的:研究微透析探针对青藤碱体外回收率的影响因素和体内回收率的稳定性,为利用微透析技术进一步研究青藤碱关节腔局部的药动学提供依据。方法:采用液质联用技术(LC-MS)测定微透析样品浓度,运用增量法考察不同药物浓度、不同灌流速度对青藤碱体外回收率的影响,运用减量法考察青藤碱体内回收率在24 h内的稳定性。结果:所建立的LC-MS分析方法准确、灵敏、可靠,可用于青藤碱在微透析样品中的含量测定;微透析探针对青藤碱的体外回收率与灌流速度成反比,与药物浓度无关;在24 h内微透析探针对青藤碱的体内回收率基本稳定。结论:可以运用微透析技术进一步研究青藤碱关节腔局部药动学。 Objiective:The influence factors of the recovery rate of sinomenine in vitro and the stability of the recovery rate in vivo were studied by microdialysis probes,which provided the basis for the further study of the pharmacokinetics of sinomenine in joint cavity by microdialysis.Methods:The concentration of microdialysis samples was determined by lc-ms.The effects of different drug concentrations and different perfusion rates on the recovery rate of sinomenine in vitro were investigated by incremental method,and the stability of the recovery rate of sinomenine in vivo within 24 h was investigated by reducing method.Results:The established LC-MS analysis method is accurate,sensitive and reliable,which can be used for the determination of sinomenine in microdialysis samples.The in vitro recovery of sinomenine by microdialysis probe was inversely proportional to the irrigation velocity and independent of the drug concentration.The in vivo recovery of sinomenine by microdialysis probe was basically stable within 24 h.Conclusion:The local pharmacokinetics of sinomenine joint cavity can be further studied by microdialysis.
