摘要
目的:采用柱色谱-紫外分光光度法测定关黄柏中总生物碱的含量,为关黄柏药材的全面质量控制提供一定依据。方法:用柱色谱-紫外分光光度法,以盐酸小檗碱为对照品,采用盐酸-乙醇(1︰100)浸渍后超声提取,经过氧化铝柱分离纯化,在265 nm处测定关黄柏中总生物碱的含量。结果:盐酸小檗碱浓度在2.36~11.80μg/mL时具有良好的线性关系,平均回收率为98.80%,重复性试验RSD为2.8%(n=5)。结论:柱色谱-紫外分光光度法测定关黄柏中总生物碱含量,方法简便,重现性好,可以作为关黄柏药材的质量控制方法之一。
Objective:To establish column chromatography-UV spectrophotometry for determination of total alkaloids(berberine hydrochloride)in Phellodendron amurense Rupr.,to provide a scientific basis for the quality control of Radix Phellodendron amurense Rupr.Methods:The sample was extracted with HCl-C2 H5 OH(1︰100)by cold after ultrasonic extraction and total alkaloids were separated on alumina column.Using berberine hydrochloride as control,the content of total alkaloids were determined by UV-spectrophotometry at detection wavelength of 265 nm.Results:The calibration curves for berberine hydrochloride was linear in the range of 2.36~11.80μg/mL.The average recovery was 98.80%,and the RSD was 2.8%(n=5).Conclusion:The method is rapid,accurate and reliable,which can be used for the quality control of Phellodendron amurense Rupr.
