雪峰虫草多糖的抗氧化活性研究 被引量:1

Study on Antioxidant Activity of Extracellular and Intracellular Polysaccharides of Ophiocordyceps Xuefengensis

摘要 为探索雪峰虫草多糖的抗氧化活性,采用邻二氮菲-Fe2+氧化法、DPPH自由基分析法、邻苯三酚自氧化法分别考察了雪峰虫草胞内多糖、胞外多糖对羟基自由基(·OH)、DPPH自由基(DPPH·)和超氧阴离子自由基(O2-·)的清除活性,并采用红外光谱分析法对其结构进行初步分析。结果表明:雪峰虫草胞内多糖和胞外多糖均表现出对3种自由基的清除活性,结果显示雪峰虫草胞内多糖的抗氧化活性优于胞外多糖,其对羟基自由基(·OH)、DPPH自由基(DPPH·)和超氧阴离子自由基(O2-·)的最大清除率分别为64.43%、62.70%和60.56%。红外光谱分析证实两种多糖均存在糖的特征吸收峰,且胞内多糖存在典型的甘露糖吸收峰。这些数据证实了雪峰虫草多糖的抗氧化活性,为其综合开发提供了必要参考。 In order to explore the antioxidant activity of extracellular and intracellular polysaccharides of Ophiocordyceps xuefengensis,phenanthroline Fe2+method,DPPH·assay method and pyrogallic acid self-oxidizing method were employed for analyzing·OH scavenging activity,DPPH·scavenging activity and super oxygen anion free radical scavenging activity of the two kinds of polysaccharides.Then Fourier transform infrared spectrometry was also used to identify the structural difference between extracellular and intracellular polysaccharides of Ophiocordyceps xuefengensis.The results indicated that both extracellular and intracellular polysaccharides could exhibit scavenging activity of·OH,DPPH·and O2-·,but intracellular polysaccharides showed better antioxidant activity with·OH scavenging activity of 64.43%,DPPH·scavenging activity of 62.70%and super oxygen anion free radical scavenging activity of 60.56%,respectively.Analysis of infra-red spectra suggested that both tested polysaccharides showed typical adsorption peaks belonged to polysaccharides,and intracellular polysaccharides also showed typical mannose adsorption peak.These data verified antioxidant activity of the two tested polysaccharides of Ophiocordyceps xuefengensis,which could provide a necessary reference for deep exploitation of fungal polysaccharide.
