结直肠癌患者血清微小RNA-29a-3p的表达及临床意义

Clinical Significance of Serum MicroRNA-29a-3p for Colorectal Cancer

目的测定结直肠癌(colorectal cancer,CRC)患者血清中微小RNA-29a-3p(miR-29a-3p)的表达水平,分析其临床意义.方法收集2017年02月-2019年02月就诊的65例CRC患者及65名健康体检志愿者,应用实时荧光定量PCR(qPCR)检测血清miR-29a-3p水平,分析其表达与临床特征之间的关系及用于诊断CRC的效能.结果CRC患者血清miR-29a-3p水平高于对照组(P<0.01),表达与临床分期、淋巴结转移有关,差异有统计学意义(P<0.05);与患者性别、年龄、分化程度、肿瘤部位、癌痛评分无关(P>0.05).血清miR-29a-3p诊断CRC灵敏度(Se)为72.90%,特异度(Sp)为84.80%,正确诊断指数(YI)为0.577,ROC曲线下面积(AUC)为0.817,与AUC=0.5比较,差异具有统计学意义(P<0.01),且其介于0.7~0.9,提示有一定的准确性.结论血清miR-29a-3p可能是CRC早诊、早治和评估预后的潜在生物标记物.

Objective To observe the expression characteristics of microRNA-29a-3p(miR-29a-3p)in the serum of patients with colorectal cancer(CRC)and analyze its clinical significance.Methods The expression level of serum miR-29a-3p was detected by real-time quantitative PCR(qPCR),the relationship between the expression level and clinicopathological characteristics and its efficacy in diagnosing CRC were analyzed in 65 CRC patients and 65 healthy volunteers who visited the Affiliated Suzhou Science and Technology Town Hospital of Nanjing Medical University from February 2017 to February 2019.Results Serum miR-29a-3p levels were significantly increased in the CRC group compared to healthy controls(P<0.01),and its expression level was related to clinical stage and lymph node metastasis(P<0.05),the difference was statistically significant,but not related to gender,age,differentiation degree of the patient,location and pain score(P>0.05).The sensitivity(Se)and specificity(Sp)of serum miR-29a-3p for CRC diagnosis were 72.90%,84.80%,and the correct diagnostic index(YI)were 0.577.The area under ROC curve(AUC)was 0.817,respectively,with statistically significant differences from AUC=0.5(P<0.01),and it is between 0.7 and 0.9,indicating a certain accuracy.Conclusion Serum miR-29a-3p is a potential biomarker for CRC diagnosis,treatment and prognosis assessment.
