摘要
葡萄是果树中感染病毒病最多的树种之一,侵染病毒种类多、分布广,是影响葡萄栽培过程中质量和产量的重要限制性因子之一.因此,建立高效、快速的葡萄病毒检测技术对防治葡萄病毒病十分重要.近年来,多种检测方法应用于葡萄病毒病检测,其中以基于血清学技术的ELISA和基于分子生物学技术的PCR为主.高通量测序技术(Next gener-ation sequencing technology,NGS)是近几年来发展起来的植物病毒检测手段.NGS极大降低了测序成本,实现了高通量,这使得我们可以获得整个病毒基因组的序列,从而为发现新病毒提供了可能.本文围绕葡萄病毒检测领域检测技术进行概述,重点阐述NGS用于检测葡萄新病毒的几个实例.
Grapevine is one of the most susceptible fruit trees to viruses.The large variety and wide distribution of infected viruses are key limiting factors for yield and fruit quality of grapevines.The establishment of an efficient and rapid virus detection technology is therefore especially important for the grapevine virus prevention and treatment.In recent years,various methods have been applied in the detection of grapevine viruses,among which the ELISA based on serological techniques and PCR based on molecular biology techniques are the main ones.Next generation sequencing technology(NGS)is a new technology helping a lot in the diagnosis of infective diseases in plants.It has realized a high-throughput screening,but at a much lower cost.It allows us to obtain sequences of the entire genome,opening up the possibility of discovering new viruses.This paper provides an overview of detection techniques in the field of grapevine virus detection,focusing on several examples of NGS used to deetct new viruses in grapevines.
