七氟烷保护缺氧性耳蜗损害新生鼠作用机制 被引量:1

摘要 目的观察七氟烷对新生鼠耳蜗一氧化氮合酶(NOS)表达的影响.方法斯泼累格·多雷(SD)大鼠30只,随机分为3组,每组10只,对照组持续吸入纯氧;七氟烷组持续吸入1.7%七氟烷加纯氧;窒息组持续吸入纯氧.吸入七氟烷或氧气30 min时处死母鼠.对照组和七氟烷组:迅速取出胎鼠,取出新生鼠听泡;窒息组:处死母鼠后取子宫放在37℃水浴槽20 min,取出新生鼠听泡.采用免疫组织化学方法测定螺旋器、血管纹和螺旋神经节诱生型NOS(iNOS)、内皮型NOS(eNOS)和神经元型NOS(nNOS)的表达水平.结果与对照组比较,七氟烷组除血管纹内iNOS、nNOS表达之外,其余耳蜗各部位各亚型NOS表达均下调(P<0.05);窒息组耳蜗Corti器、血管纹、螺旋神经节内各型NOS表达下调(P<0.05或P<0.01).与七氟烷组比较,窒息组耳蜗螺旋神经节内eNOS和nNOS表达下调明显(P<0.05).结论七氟烷对缺氧性耳蜗损害新生鼠有保护作用.
