细胞衰老及相关抗衰策略研究进展

摘要 细胞衰老(cellular senescence)属于细胞水平的衰老,是细胞周期阻滞的一种稳定终末状态,并引起各种衰老相关分泌表型(senescence-associated secretory phenotype,SASP);发生机制主要涉及DNA损伤、端粒功能障碍、癌基因激活、线粒体功能障碍、染色质退变以及SASP组成与调控等。细胞衰老累及包括角质形成细胞、成纤维细胞、黑素细胞及各类免疫细胞等;治疗包括senolytics策略、senomorphics策略以及调控衰老细胞恢复正常生理及分泌功能的再生技术。该文综述细胞衰老相关机制、皮肤衰老细胞变化、细胞衰老相关抗衰策略及其研究进展。 Cell senescence occurs at the cellular level,is a stable terminal state of cell cycle arrest,which causes various senescence-associated secretory phenotype(SASP).The pathogenesis mainly involves DNA damage,telomere dysfunction,proto-oncogene activation,mitochondrial dysfunction,chromatin degeneration,and SASP composition and regulation.Cell senescence involves keratinocytes,fibroblasts,melanocytes and various immune cells.The treatment includes senolytics strategy,senomorphics strategy and regeneration technology that regulates senescent cells to restore normal physiological and secretory functions.In this paper,the related mechanisms of cell senescence,changes of skin senescence cells,anti-aging strategies related to cell senescence and their research progress are reviewed.
