落叶松中二氢槲皮素的提取及稳定性 被引量:1

Extraction and Stability of Dihydroquercetin in Larch

摘要 采用浸取法,以乙二醇作为提取溶剂从落叶松中提取二氢槲皮素,通过正交试验确定最佳提取条件,采用反相高效液相色谱法测定提取液的主要成分,对从落叶松中提取的二氢槲皮素在不同环境条件下进行稳定性的研究.结果表明:正交试验确定最佳提取条件为乙二醇体积分数60%、提取温度40℃、料液比1:30(g/mL)、提取时间90min;反相高效液相色谱法测定提取液的主要成分是二氢槲皮素;固态的二氢槲皮素在217℃有吸热峰出现,但在水相中加热时间越长二氢槲皮素越不稳定,pH值对二氢槲皮素有显著的影响,金属离子Ba^(2+)、Na^(+)、Ca^(2+)、Mg^(2+)、NH^(4+)对二氢槲皮素无影响,AI^(3+)、Cu^(2+)、Fe^(3+)对其稳定性有不良影响. Dihydroquercetin,as the primary flavonoids in larch,was extracted using ethylene glycol as the extraction solvent Hie optimal extraction conditions were explored by orthogonal tests to be ethylene glycol concentration of 60%,extraction temperature of 40℃,material-liquid ratio of 1:30(g/mL)and extraction time of 90 min.The dihydroquercetin as the principal component in extract was determined by reversed-phase high-performance liquid chromatography(HPLC).The stability of dihydroquercetin was investigated under various environmental conditions.The results showed that the endothermic peak of solid dihydroquercetin was observed at 217"C.Hie dihdroquercetion is unstable in aqueous phase during the long-time heating treatment.The stability of dihydroquercetin was significantly affected by pH.However,metal ions such as Ba^(2+),Na^(+),Ca^(2+),Mg^(2+)and NH^(4+)had no obvious effect on the stability of dihydroquercetin although AI^(3+),Cu^(2+)and Fe^(3+)had negative impact on its stability.
