稻谷原粮中真菌多样性研究 被引量:3

摘要 采用形态学观察结合ITS及26S序列的鉴定来确定真菌的种属。通过系统发育树的构建来确定稻谷真菌之间的亲缘关系及多样性。通过形态学观察结合真菌ITS序列及26S序列分别对稻谷原粮中分离获得的33株真菌进行了鉴定,主要为青霉菌(Penicillium),曲霉属(Aspergillus),镰刀菌(Fusarium)等菌属,其中青霉属与曲霉属最多分别为16株,15株,而镰刀菌属仅2株。两种序列鉴定结果的结合能更加准确地鉴定稻谷原粮中菌株的种属,为稻谷储藏过程中的真菌种类鉴别及生物控制提供研究基础。 Fungal strains in raw rice had been isolated and identified with the sequence of 26S rDNA and ITS adopted in this research,based on the sequences the genus of the fungi.Method:With morphological characterization,and rDNA-ITS analysis,the species of fungal in raw rice were identified and differentiated.By means of the multiple comparison analysis of ITS sequences and 26S rDNA sequences,the diversity ofintra-species in the rice Fungal were investigated.Result:Three strains were identified,e.g.Penicillium,Aspergillus,Fusarium,Penicillium was the dominant genus,accounting for 48%of the tested strains.And Aspergillus was the second dominant species,accounting for 45%.Only two Fusarium strains were found in this research.Conclusion:Thirty-three strains were isolated and identified through morphology and molecular biology.With 26SrDNA and ITS sequences analysis can show a more accurate result,which can provide foundation for biological control in rice Spoilage mold.
