The research about effect of Zizhu Ointment on autophagy proteins LC3 and Becin1 during acute wound healing

摘要 Objective: To observe the effect of Zizhu ointment on acute wound healing and its effect on the expression of autophagy proteins LC3 and Becin1. Method: SD rat model with acute skin injury were randomly divided into groups. The experimental group was treated with external application of Zizhu ointment, and the control group was given external saline. The wound healing rate of the two groups was compared at 4d, 7d, 14d and 21d after trauma. The morphology of wounds was observed by HE staining, and the autophagosomes of wounds were observed by transmission electron microscopy. Expression and changes of autophagy marker proteins Beclin-1 and LC3 were detected by immunohistochemistry and WB assay. Result: Compared with the control group, experimental group had a high healing rate, a short healing time, and a faster epithelialization rate. Under the electron microscope, autophagosomes of experimental group were reduced, and the expression of LC3 and Beclin-1 was significantly decreased. Conclusion: Zizhu ointment may promote wound healing by inhibiting autophagy activity of cells.
