摘要
目的 分析电磁辐射暴露对妊娠大鼠氧化应激、炎性反应和子代的影响.方法 选取24只清洁级Wistar妊娠期大鼠,采用随机数字表法分为对照组(假暴露,不接受辐射)、低剂量组(10 min/次)、中剂量组(60 min/次)、高剂量组(120 min/次),每组6只,孕鼠自妊娠第1天暴露于1800 MHz电磁辐射,3次/d,直至仔鼠出生,测定孕鼠分娩时氧化应激指标[超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、脂质过氧化物(MDA)]、炎性反应因子(TNF-α、IL-6、IL-1),同时在仔鼠出生后1个月进行Morris水迷宫试验,观察仔鼠学习记忆能力,并记录子代生长发育情况(体质量、身长及尾长).结果 中、高剂量组SOD、GSH-Px、CAT活性低于其他各组,而MDA活性、血清TNF-α、IL-6、IL-1水平高于其他各组,且低剂量组、中剂量组、对照组上述指标差异也有统计学意义(P<0.05);Morris水迷宫试验发现高剂量组仔鼠过台次数少于其他各组(P<0.05).结论 电磁辐射暴露可引起妊娠大鼠氧化应激及炎性反应,同时可能对其子代学习记忆能力造成不良影响.
Objective To determine the effects of electromagnetic radiation exposure on oxidative stress and inflammatory response in pregnant rats and offspring.Methods Twenty-four clean-grade Wistar pregnant rats were included and randomly divided into control group(sham exposure,no radiation),low-dose group(10 min/time),medium-dose group(60 min/time)and high-dose group(120 min/time),with 6 rats in each group.The pregnant rats were exposed to 1800MHz electromagnetic radiation from the first day of pregnancy for 3 times/d until the birth of offspring.The oxidative stress indicators[superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)and lipid peroxide(MDA)]and inflammatory response factors[tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-1(IL-1)]were measured in the pregnant rats at delivery.Meanwhile,Morris water maze test was performed at 1 month after the birth of offspring.The learning and memory abilities of the offspring were determined,and the growth and development(weight,height and tail length)were recorded.Results The activities of SOD,GSH-Px and CAT in the medium-and high-dose groups were lower than those in the other groups,whereas the MDA activity and the levels of serum TNF-α,IL-6 and IL-1 were higher than those in the other groups.There were statistically significant differences in the above-mentioned indicators between the low-dose group,medium-dose group and control group(P<0.05).The Morris water maze test found that platform-crossing times in the high-dose group was lower than that in the other groups(P<0.05).Conclusion Electromagnetic radiation exposure may cause oxidative stress and inflammatory response in pregnant rats,and may bring adverse effects on the learning and memory abilities of their offspring.
