食源性相关腹泻患者粪弯曲菌检测结果分析

目的 探讨多重聚合酶链式反应(PCR)技术与增菌培养分离鉴定法检测食源性相关腹泻患者粪弯曲菌的效果和可行性.方法 比较多重PCR技术与增菌培养分离鉴定法对200例食源性相关腹泻患者粪弯曲菌的检测,在培养鉴定中使用VITEK-2?compact全自动微生物分析系统(法国bioMérieux公司)的苛养菌配套鉴定卡,并相应增加单重、二重或多重PCR的检测.结果 多重PCR的检出率为2.50% (5/200);而增菌培养分离法的检出率为2.00%(4/200),有1例未能分离出菌落但平板盲刮PCR仍可检出;多重PCR的检出空肠弯曲菌4例,结肠弯曲菌1例;增菌培养分离法菌种生化鉴定菌种结果与多重PCR一致.结论 多重PCR技术比较适合于食源性相关腹泻患者粪弯曲菌的快速检测,也应重视培养分离法对鉴定与药敏的地位,努力优化选择性培养基及培养条件.

Objective To explore the relationship between detecting fecal Campylobacter bymultiplex PCR technology and the results of enrichment culture isolation in patients with foodborne diarrhea.Methods Fecal Campylobacter in 200 patients with foodborne diarrhea were detected by multiplex PCR and an enrichment culture isolation method.VITEK-2 compact automatic microbial analysis system(Frence bioMérieux company)and its the fastidious bacteria identification card were matching in the cultivation and identification,and to increase the detection of single,double or multiple PCR accordingly.Results The detection rate of multiple PCR was 2.50%(5/200).And the detection rate of enrichment culture isolation method was 2.00%(4/200),1 case failed to isolate the colony,but was still detected by all the PCR.4 cases of Campylobacter jejuni and 1 cases of Campylobacter coli were detected by multiplex PCR.The results of strain biochemical identification by enrichment culture and isolation were consistent with multiple PCR.Strain identification by enrichment culture were consistent with multiple PCR.Conclusion Multiplex PCR is more suitable for rapid detection of fecal Campylobacter in foodborne diarrhea patients.We should also attach importance to the cultivation of separation method for identification and drug sensitivity,and try hard to optimize selective medium and culture conditions.
