微生物实验室常用菌种保存方法及临床意义的探讨

摘要 目的 探索微生物实验室常用菌种的适宜保存方法,为临床提供准确可靠的检验结果.方法 采用6种菌种保存方法,对22种常用菌种进行保存,探究各种方法下菌种存活率和最佳保存时间.结果 血琼脂培养基低温保存法、固体培养基低温保存法和营养琼脂斜面培养基低温保存法,第4个月菌种存活率下降至<50%,甚至降为0%;甘油肉汤法和液体石蜡法,第6个月菌种存活率>70%;脱脂牛奶法第18个月菌种存活率为100%,并且菌量和菌落形态正常,生化反应特性基本不变.结论 血琼脂培养基低温保存法、固体培养基低温保存法和营养琼脂斜面培养基低温保存法适合菌种短期内保存;甘油肉汤法和液体石蜡法适合菌种长期保存;脱脂牛奶低温冻存法则可以将菌种进行超长期保存. Objective To explore appropriate preserve methods of common bacteria strains in microbiology laboratory in order to provide accurate and reliable test result for clinical departments.Methods Six methods of preserving bacteria strains were used to storage 22 kinds of common bacteria strains in order to explore the survival rates and optimal preserve time of common bacteria strains.Results Survival rates of bacterial strains were down to less than 50%with the cryopreservation methods of blood agar medium,solid medium and nutrition agar inclined medium in the forth month,even down to 0%,and the survival rates were more than 70%of glycerol broth method and liquid paraffin method in the sixth month,preservated with skimed milk method,the survival rate was 100%in the eighteenth month with the amount,colony morphology,biochemical features basically remain unchanged.Conclusion The cryopreservation methods of blood agar medium,solid medium and nutrition agar inclined medium are suitable for the short term preservation of bacterial strains,while glycerol broth method and liquid paraffin method are suitable for the long-term preservation,what is more,skimed milk cryopreservation method is suitable for a ultra-long-term preservation.
