前列地尔对大鼠超长皮瓣存活影响的实验研究

目的 探讨前列地尔对大鼠超长任意皮瓣存活面积的影响.方法选取(250±30)g?SD大鼠40只,随机分为观察组和对照组,各20只,每只大鼠均在腹部行超长任意皮瓣转移术,观察组在大鼠腹腔内注射10ml/(kg·d)的前列地尔稀释液,对照组在大鼠腹腔内注射10ml/(kg·d)生理盐水;7d后分别检测两组皮瓣存活面积,使用ELISA法检测每只大鼠血中VEGF、IL-1β及TGF-β1的含量,病理切片观察皮瓣血管再生情况.结果 术后7d,观察组皮瓣存活面积显著高于对照组,观察组VEGF、TGF-β值均高于对照组,IL-1β则低于对照组,差异均有统计学意义(P<0.05);观察组组织炎症、皮瓣水肿、血管再生等情况也优于对照组.结论 前列地尔能够减轻皮瓣移植术后组织水肿,改善静脉瘀血,减轻皮瓣组织炎症、促进毛细血管新生,显著提高大鼠腹部皮瓣转移的成功率.

Objective To study the effect and mechanism of alprostadil on the survival area of rat ultra-long random flaps.Methods A total of 40 250±30g SD rats were randomly divided into observation group and control.Each rat underwent ultra-long flap transfer in the abdomen.The observation group were injected with 10 ml/kg/d of alprostadil diluent in the abdominal cavity of the rats,and the control group was injected with 10ml/kg/d normal saline in the abdominal cavity of the rats.The survival area ratio of the two groups was measured after 7 days,and the contents of VEGF,IL-1βand TGF-β1 in the blood of each rat were detected by ELISA.The angiogenesis of the flap was observed by pathological section.Results After 7 days,the survival area of the flaps in the observation group was significantly higher than that in the control group.The VEGF and TGF-βvalues in the observation group were higher than those in the control group,and IL-1βwas lower than the control group.The difference was statistically significant(P<0.05).Tissue inflammation,flap edema,and angiogenesis were also better in the observation group than in the control group.Conclusion Alprostadil can reduce tissue edema after flap transplantation,improve venous congestion,reduce inflammation of flap tissue,promote capillary angiogenesis,and significantly improve the success rate of abdominal flap transfer in rats.
