七氟烷预处理后对大鼠脑缺血的神经保护作用

摘要 目的 观察七氟烷预处理对大鼠缺血性脑损伤的神经保护作用.方法 将SD大鼠分为实验组、对照组和假手术组,七氟烷预处理实验组大鼠后,实验组和对照组建立局灶性脑缺血再灌注模型(MCAO),24 h后各组进行神经功能缺损评估、测量脑梗死面积,MCAO 3d后TUNEL法检测细胞凋亡.结果 实验组神经功能缺损评分明显低于对照组(<0.05),实验组脑缺血损伤面积明显小于对照组(<0.05),MCAO 3d后实验组TUNEL阳性细胞明显少于对照组(<0.05).结论 七氟烷预处理对于缺血性脑损伤有神经保护作用. Objective To investigate the neuroprotection of sevoflurane preconditioning for local cerebral ischemia.Methods 36 two-month old Sprague-Dawley male rats were randomly divided into sham group,experimental group and control group.Experimental group was exposed to sevoflurane for 30 min/day on 4 consecutive days and then underwent a 60 min middle cerebral arterial occlusion(MCAO),while control group underwent MCAO only.At 24 h after reperfusion all the groups were investigated by neurological severity score(NSS)for neurological deficits,TTC staining for infarct percentage,TUNEL was performed to determine the percentage of cell death after MCAO 3d.Results The NSS score and infarct percentage in experimental group decreased significantly compared with those of control group at 24 h after reperfusion(<0.05),The percentage of TUNEL positive cells of experimental group was reduced significantly compared to that of control group at 3 d after reperfusion(<0.05).Conclusion Sevoflurane preconditioning provides neuroprotection against local cerebral ischemia.
