HPLC测定痛泻宁颗粒中芍药苷的含量

Determination of Paeoniflorin in Tongxiening Granule by HPLC

目的:本实验为了建立痛泻宁颗粒中芍药苷的测定方法。方法采用高效液相色谱法测定,采用依利特SinoChrom ODS-BP(250 mm×4.6 mm,5μm);流动相:乙腈-0.1%磷酸(15∶85);体积流量:1.0 ml?min-1;检测波长:230 nm;柱温:25℃;进样量10μl。结果芍药苷进样量在0.248~2.480μg呈良好的线性关系(=0.9998),平均回收率为98.52%,RSD值为1.43%(n=6)。结论实验表明本研究方法专属性强,灵敏度高,简便、准确、重现性好,可用于痛泻宁颗粒中芍药苷的测定。

Objective To establish a method for the determination of paeoniflorin in Tongxiening Granule.Methods The content of paeoniflorin was determined by HPLC,Elite SinoChrom ODS-BP column(250 mm×4.6 mm,5μm)was used with acetonitrile-0.1%orthophosphoric acid(15∶85)as the mobile phase at the flow rate of 1.0 mL·min-1.The detection wavelength was set at 230 nm and the column temperature was 25℃.The injection volume was 10μl.Results The linear range of paeoniflorin was 0.248~2.480μg(r=0.999 8)with an average recovery of 98.52%(RSD=1.43%,n=6).Conclusion The established method is specific,sensitive,simple,accurate,reproducible,and suitable for the quality control of paeoniflorin in Tongxiening Granule.
