乙肝宁颗粒抗乙型肝炎病毒的体外实验研究

Inhibitory Effect of YiGanNing Granules on HepatitisB Virus in Vitro

目的:探讨体外乙肝宁颗粒抗乙肝病毒活性。方法将不同浓度乙肝宁颗粒与HepG硬硬15细胞株共培养9d,以拉米夫定为阳性对照药,以MTT比色法观察药物细胞毒性;用ELIS法测定细胞上清液中乙型肝炎病毒e抗原( HBeAg),乙型肝炎病毒s抗原( HBsAg);采用荧光定量PCR法(FQ-PCR)测定细胞上清液HBV DNA,计算抑制率。结果乙肝宁颗粒在体外能有效抑制HepG硬硬15细胞分泌HBsAg和HBeAg,最大抑制率分别为54.69%和25.93%;对HBV DNA的复制有一定抑制作用(<0.05)。结论乙肝宁颗粒能有效抑制HepG硬硬15细胞中HBV复制,说明在体外具有显著的抗乙型肝炎病毒作用,且毒性较小,具有良好应用前景。

Objective To observe the antiviral activities of YiGanNing Granules on hepatitis B virus(HBV)in vitro.Methods HepG⒉⒉15 cells were treated with dif erent concentrations(25,50,100,200,400mg·L-1)of YiGanNing Granules for 9d.Cytotoxicity was observed with MTT colorimetric method.HBeAg and HBsAg in the culture supernatant were determined by ELISA assay.Fluorescence quantity PCR assay was used to assay the extracellular HBV DNA.Results After treatment with 400 mg·L-1 YiGanNing Granules for 9d the level of HBeAg and HBsAg in culture supernatant.When OM concentration was 400 mg·L-1,the inhibitory rates on HBsAg and HBeAg were54.69%and 25.93%.It can also inhibit the level of the extracellular HBV DNA(<0.05)and the inhibitory ef ect showed a dose-and time-dependent manner.Conclusion YiGanNing Granules can ef ectiveliy inhibit HBV DNA replication in HepG⒉⒉15 cells in vitro with low toxicity,which shows a fine applying prospect.
