脑损伤后孕酮促进神经功能的分子机制

摘要 目的:观察孕酮对大鼠脑损伤后iNOS表达及神经功能的影响。方法雄性 SD大鼠随机分为假手术组、脑损伤组和孕酮治疗组,按照改良的 Feeney自由落体损伤装置制作大鼠脑损伤模型。孕酮治疗组伤后1h、6h腹腔注射孕酮16 mg/kg。各组于伤后24h取材。用免疫组化法检测 iNOS含量,用 mNSS评分检测神经功能。结果孕酮治疗组与脑损伤组比较, iNOS表达明显减少,神经功能明显改善,差异有统计学意义(P<0.05)。结论孕酮可能通过降低 iNOS的表达,改善神经功能,发挥脑损伤保护作用。 Objective To investigate ef ects of progesterone treatment on iNOS and neurological function after traumatic brain injury in rats.Methods Male Spraque-Dawley rats wererandomly divided into 3 groups:sham-operated group,TBI group and PROG-treated group.The rat model of TBI was duplicated with the improved Feeney's method.The injections of progesterone in the PROG-treated group were given i.p.at 1h and 6h after the injury.The rats were kil ed respectively in three groups at 24h after injury.iNOS were detected using immunohistochemisty.The neurological outcome was assessed by Modied neurological severity score(mNSS)test.Results Progesterone treatment improves neurological function,decreases the expression of iNOS after traumatic brain injury.There were significant dif erences between the TBI group and the PROG-treated group(P<0.05).Conclusion The protective ef ect of progesterone on neurological function may be induced by down-regulation of iNOS expression after traumatic brain injury.
