摘要
基于多克隆抗体建立了烯效唑的时间分辨荧光免疫分析(TRFIA)方法。采用烯效唑半抗原与卵清白蛋白(OVA)的偶联物包被96孔板为固相抗原,与样品中游离的烯效唑共同竞争Eu3+标记的抗烯效唑抗体,用直接竞争法检测样品中的烯效唑,在水和土壤中分别添加烯效唑标准溶液,平均回收率为80.4%~116.8%,变异系数为2.6%~8.7%,符合农药残留检测要求,该TRFIA方法可用于环境与食品中烯效唑残留高灵敏度检测。
A sensitive time-resolved fluoroimmunoassay(TRFIA)based on polyclonal antibody for detection of uniconazole was developed.A hapten was synthesized and conjugated with ovalbumin(OVA)as the coating antigen.The coating antigen was coated by physical adsorption onto a 96 wel microtitre plate,which was then competed with uniconazole in the sample for limited quantity of anti-uniconazole antibody labeled by Eu3+.Uniconazole in the samples could be determined by direct competition method.At spiked dif erent levels(0.05~0.5 mg/kg in water and soil)recoveries of uniconazole were from 80.4%~116.8%,with CV of 2.6%~8.7%.The TRFIA could be a convenient tool for monitoring uniconazole residues in environment and food samples.
